High mobility group box proterin-1 (HMGB-1) is a multifunctional protein that can be released by various programmed cell deaths (PCDs), such as necroptosis and ferroptosis. PCDs play critical role in the pathogenesis of systemic lupus erythematosus (SLE). However, HMGB-1 process SLE remains unclear. This study aims to demonstrate potential diagnosing serum We found levels HMGB-1, receptor-interacting kinase 3 (RIPK3) /mixed lineage domain-like (MLKL) related with necroptosis, metabolites associated ferroptosis were significantly upregulated patients compared HC individuals. These positively correlated disease activity. Additionally, level showed strong positive RIPK3/MLKL metabolites. Moreover, was renal involvement high-antinuclear antibodies (ANA) titer. After interferon γ (IFN-γ) treatment vitro, markers activated HMGB1 both HEK293 HK2 cells. Clinically, considered significant independent risk factor binary logistic assay. Notably, exhibited outstanding diagnostic ability for area under curve (AUC) receiver operating characteristic (ROC) analysis. Taken together, our indicates promising biomarker diagnosis monitoring SLE.

The non-high-density lipoprotein cholesterol to high-density ratio (NHHR) is a newly developed lipid parameter. However, the current research has only explored relationship with lumbar spine bone mineral density, lacking studies on density at other sites, total body and an analysis of risk factors. This study aims determine potential association between NHHR BMD, increase awareness impact levels health. By utilizing data from National Health Nutrition Examination Survey (NHANES) 2011 2018, we conducted univariate generalized linear models (GLMs) analysis, stratified threshold effect smooth curve fitting investigate BMD. were categorized into tertiles (low, medium, high) based their distribution among population. included 8,671participants, have shown, exhibits correlation (BMD). In BMI subgroup, significantly negatively correlated BMD multiple sites in low-to-middle group (BMI <25 kg/m2), while no significant found high ≥30 kg/m2). gender more pronounced male mainly reflected reduction age negative strongest younger (18-30 years), gradually weakening middle-aged (31-44 years) older groups (45-59 years). Further suggests that dyslipidemia may influence metabolism through pathways such as inflammation oxidative stress. (BMD) varies by BMI, gender, age. controlling be intervention target for health management, particularly individuals males, populations. These findings offer new perspective provide scientific evidence development personalized osteoporosis prevention treatment strategies.

Metabolic diseases (MDs), exemplified by diabetes, hypertension, and dyslipidemia, have become increasingly prevalent with rising living standards, posing significant public health challenges. The MDs are influenced a complex interplay of genetic factors, lifestyle choices, socioeconomic conditions. Additionally, environmental pollutants, particularly air pollutants (APs), attracted increasing attention for their potential role in exacerbating these MDs. However, the impact APs on remains unclear. This study introduces novel machine learning (ML) pipeline, an Algorithm Spatial Relationships Analysis between Exposome Diseases (ASEMD), to analyze spatial associations at prefecture-level city scale China. ASEMD pipeline comprises three main steps: (i) autocorrelation is evaluated using Moran's I statistic Local Indicators Association (LISA) maps. (ii) dimensionality reduction similarities identification clusters Principal Component (PCA), k-means clustering, Jaccard index calculations, further validated through (iii) AP exposure adjusted demographic confounders predict models (e.g., eXtreme Gradient Boosting (XGBoost), Random Forest (RF), Decision Tree (DT), LightGBM, Multi-Layer Perceptron (MLP)). Objectives Hepatic osteodystrophy (HOD) is an important public health issue that severely affects human health. The pathogenesis of HOD complex, and exposure to environmental pollutants plays role. Di-(2-ethylhexyl) phthalate (DEHP) a persistent endocrine toxicant present in many products, the liver target organ for its toxic effects. Our research aimed investigate effects DEHP on HOD, reveal underlying mechanisms potential key preventive approaches. Methods daily intake EDI bone density indicators men women from 2009 2018 were screened organized NHANES database population correlation between BMD; C57BL/6 female male mice selected construct animal model induced exploring fuchtions osteoporosis; novel small molecule inhibitor imICA was used inhibit process osteoporosis, further targeted inhibition pathway HOD. Results Male populations exposed relatively lower concentration DEHP, only exhibited negative mineral density. An vivo study confirmed low dose caused lesions, disrupted function, osteoporosis but not C57BL/6J mice. Abstract Osteoporosis is classified as a metabolic syndrome, and the consumption of fructose has been linked to various diseases. However, specific effects underlying mechanisms on bone health remain inadequately understood. In this study, we demonstrate that intake can exacerbate loss in murine models by facilitating accumulation cholesterol within bones. We identify Thyroid Hormone Receptor Beta (Thrb) Protein Kinase C Zeta (Prkcz) potential therapeutic targets for treatment osteoporosis. Mice subjected high-fructose diet exhibited reduction density decrease osteogenic differentiation marrow mesenchymal stem cells (BMSCs) compared those standard diet. Fructose was found Thrb expression while increasing Prkcz expression, leading hindering BMSCs. Furthermore, our findings indicate activation inhibition significantly ameliorate mice. This study elucidates molecular which influences osteogenesis through Thrb/Prkcz/cholesterol pathway context osteoporosis, thereby highlighting osteoporosis treatment.
