Echinacoside (ECH) is a prominent naturally occurring bioactive compound with effects of alleviating myocardial damage. We aimed to explore the beneficial ECH against sepsis-induced damage and elucidate potential mechanism. Echocardiography Masson staining demonstrated that alleviates cardiac function fibrosis in cecal ligation puncture (CLP) model. Transcriptome profiling network pharmacology analysis showed there are 51 overlapping targets between ECH. Subsequently, chemical carcinogenesis-reactive oxygen species (ROS) were enriched multiple targets. Wherein, SOD2 may be target on Polymerase chain reaction results administration could markedly increase expression reduce release ROS. Combined injecting inhibitor SOD2, effect mortality, function, was eliminated, ROS increased after inhibiting SOD2. significantly alleviated septic mice, therapeutic mechanism achieved by upregulating which decreased

Sepsis-induced myocardial injury (SIMI) is a vital pathological component of severe sepsis and septic shock. As prevalent internal mRNA modification in eukaryotic cells, N6-methyladenosine (m6A) implicated immune disorders. Methyltransferase-like 14 (METTL14), core subunit the methyltransferase complex that catalyzes messenger RNA m6A modification, involved regulation human cardiomyocyte cell line (AC16) injury. This study aimed to explore role mechanism METTL14 lipopolysaccharide (LPS) -induced injury.Cell viability apoptosis were analyzed via 3- (4,5-Dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide (MTT), flow cytometry, TdT-mediated dUTP nick-end labeling (TUNEL) assay. The Tumor necrosis factor alpha (TNF-α) Interleukin-1beta (IL-1β) levels Enzyme linked immunosorbent assay (ELISA). Caspase-3 activity, reactive oxygen species malondialdehyde level, glutathione level assessed using special kits. transient receptor potential melastatin 7 (TRPM7) determined Real-time quantitative polymerase chain reaction (RT-qPCR). Meanwhile, protein TRPM7, METTL14, phospho-p65 (p-p65), total p65 (p65), p-IκBα, IκBα (IκBα) examined western blot assay.LPS treatment repressed AC16 induced apoptosis, inflammatory response, oxidative stress, ferroptosis vitro. TRPM7 upregulated LPS-treated cells. At molecular could increase stability methylation. Moreover, deficiency abolish LPS-triggered regulation.METTL14 knockdown reversed LPS-caused damage mainly by regulating mRNA, providing novel therapeutic target for cardiomyopathy treatment.

Amaç: Mediastinit; nadir görülen ciddi bir enfeksiyondur. Bu çalışmanın amacı, post-sternotomi mediastinit (PSM) veya descending nekrotizan (DNM) tanısı alan hastaların demografik, klinik ve laboratuvar özelliklerini karşılaştırmak mortalite belirlemektir. Gereç Yöntemler: çalışmaya 2015-2022 yılları arasında Sağlık Bilimleri Üniversitesi Dışkapı Yıldırım Beyazıt Eğitim Araştırma Hastanesi'nde PSM DNM hastalar dahil edilmiştir. Hastalar hayatta kalanlar kalamayanlar olarak kategorize edilmiştir karşılaştırılmıştır. Bulgular: konulan 25 hasta Ortalama yaş 54.9 ± 12.1 olup, %64'ü erkekti. Hastaların %92'sinden kan kültürü alınmıştır. %88'inden pürülan akıntı kültürleri gönderilmiştir, %44'ünde üreme tespit %84'ünde komorbidite mevcuttur varlığı, sepsis varlığı tanı sonrası yoğunbakım ünitesinde (YBÜ) yatış günü prevalansı hastalarında anlamlı daha yüksekti (p=0.017, p=0.004, p=0.026). Kalp yetmezliği, koroner arter hastalığı (KAH) hipertansiyon (HT) (ps=0.000). hastaları; kalamayan grupta da yaygındı (p=0.012). Hayatta ortalaması yüksek, sigara içenlerin sayısı fazla YBÜ'de kalma süresi uzundu (p=0,046, p=0,049, p=0,038). PSM, HT KAH olan (p=0.012, p=0.008, p=0.033). Sonuç: ancak yüksek morbidite oranlarına sahip Yaşlılar, içenler, median sternotomi yapılanlar komorbiditeleri olanlar gibi riski hastalarda tedavi takip stratejileri geliştirilebilir.
