RNA regulation plays an integral role in tuning gene expression and is controlled by thousands of RNA-binding proteins (RBPs). We develop use a high-throughput recruitment assay (HT-RNA-Recruit) to identify regulatory domains within human RBPs recruiting over 30,000 protein tiles from 367 reporter mRNA. discover 100 unique RNA-regulatory effectors 86 distinct RBPs, presenting evidence that contain functionally separable dictate their post-transcriptional control expression, some with activity at 5' or 3'UTRs. downregulate both when recruited DNA RNA, dissect mechanisms regulation. Finally, we build synthetic regulator can stably maintain desired levels are predictable mathematical model. This work serves as resource for expands the repertoire RNA-based genetic tools.
