Background: Human papillomavirus (HPV) is a key driver of cervical cancer, with high-risk genotypes, mainly HPV 16 and 18, responsible for approximately 70% cases. Persistent infections immunological evasion by increase the risk malignant transformation. Objective: To evaluate effect TaqMan probes in real-time PCR pivotal role DNA detection, targeting highly conserved regions like E7 gene. Methods: A case-control study was conducted on 71 women Hilla City, Iraq, to detect DNA. The used swabs blood samples, Statistical analysis, including chi-square tests, evaluated associations between demographic, clinical, behavioral variables. Results: Out 22 tested positive HPV, primarily using (95.5%). Amplification gene showed high efficiency (100.9%) specificity. Significant were found infection factors such as smoking, rural residence, family history cancers. No co-infections observed. Conclusions: This underscores effectiveness probe-based RT-PCR detection highlights critical demographic factors. findings advocate expanding vaccination diagnostic access, particularly underserved regions.
