To investigate the diagnostic value of peripheral blood lymphocyte testing in children with infectious mononucleosis (IM). A total 135 IM as group and 100 healthy volunteers were included this retrospective study. Peripheral subsets marked CD3+, CD4+, CD8+, CD16 + CD56+, CD19 quantified using flow cytometry. Statistical analysis was performed chi-square test, Kruskal-Wallis AUROC curve, Kappa consistency test to assess these markers IM. The curve for CD8 cells CD4+/CD8 ratios both achieved a 1 sensitivity specificity 100% (P<0.001). coefficients combined EBV analysis, indicating clinical diagnosis. Significant differences also observed between (P<0.05). evaluation lymphocytes represents significant advancement diagnosis offers reliable, sensitive, specific tool enhance management
