Chronic rheumatic diseases such as rheumatoid arthritis (RA) are characterized by a dysregulated immune response and persistent inflammation. The large number of neutrophilic granulocytes in the synovial fluid (SF) from RA patients leads to elevated enzyme activities, for example, myeloperoxidase (MPO) elastase. Hypochlorous acid (HOCl), most important MPO-derived product, is strong reactive oxygen species (ROS) known be involved processes cartilage destruction (particularly regarding glycosaminoglycans). Abstract Purpose This study aimed to develop a transdermal delivery system for fluocinolone acetonide (FLA), corticosteroid used in treating inflammatory conditions like rheumatoid arthritis (RA), overcome the limitations of oral administration, such as poor solubility and bioavailability. Methods FLA-loaded PEG decorated hyalurosomes (FLA-PHs) were fabricated using ethanol injection, incorporating various Brij® surfactants different amounts hyaluronic acid (HA) based on full factorial design. The impact independent variables, HA amount (mg) (X 1 ) Brij type 2 inspected entrapment efficiency (EE%), particle size (PS), zeta potential (ZP). optimum FLA-PHs then incorporated into ε-polycaprolactone (PCL) cellulose acetate (CA) nanofibers enhance sustained (FLA-NFs). Results exhibited EE% 83.58 ± 0.69%, PS 169.00 1.41 nm, ZP -22.90 0.14 mV. Morphological assessment FLA-NFs showed promising results terms surface roughness. In Freund-induced rat model adjuvant-induced arthritis, treatment with significantly improved joint histopathological analyses. Furthermore, it suppressed markers mTORC1, TNF-α, NF-κB while upregulating TRIM24 anti-inflammatory IL-10. Conclusion present strategy enhancing FLA managing RA, offering improvements efficacy reduced systemic side effects compared conventional administration.

Periodontal disease, a prevalent oral inflammatory condition, is implicated in exacerbating systemic diseases. However, the molecular mechanisms underlying this association remain unclear. In study, we performed RNA sequencing of gingival tissue samples collected from mouse model periodontal disease at multiple time points to investigate dynamic transcriptomic changes during progression. Our analysis revealed distinct temporal gene expression patterns associated with key and immune response pathways. These findings suggest stepwise progression process, potentially contributing inflammation through shared signaling networks. We further identified specific genes pathways that may mediate bidirectional relationship between conditions such as cardiovascular diabetes. Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent inflammation and progressive joint destruction. Recent data underscore oxidative stress as primary factor in the pathophysiology of rheumatoid arthritis, intensifying inflammatory processes tissue damage via overproduction reactive oxygen species (ROS) compromised antioxidant defenses. Current therapies, including disease-modifying antirheumatic drugs (DMARDs), primarily target immune dysregulation but fail to address stress, necessitating novel adjunctive treatment strategies. This review explores potential antioxidant-based therapies complementary approaches RA management. Natural compounds such curcumin, resveratrol, sulforaphane, propolis exhibit strong anti-inflammatory antioxidative properties modulating redox-sensitive pathways, nuclear kappa-light-chain-enhancer activated B cells (NF-κB) erythroid 2-related 2(Nrf2)/heme oxygenase (HO-1). N-acetylcysteine (NAC) replenishes intracellular glutathione, enhancing cellular resilience against stress. Additionally, molecular hydrogen (H2) selectively neutralizes harmful ROS, reducing inflammation.
