Aging
populations
worldwide
are
placing
age-related
diseases
at
the
forefront
of
research
agenda.
The
therapeutic
potential
natural
substances,
especially
propolis
and
its
components,
has
led
to
these
products
being
promising
agents
for
alleviating
several
cellular
molecular-level
changes
associated
with
diseases.
With
this
in
mind,
scientists
have
introduced
a
contextual
framework
guide
future
aging
research,
called
hallmarks
aging.
This
encompasses
various
mechanisms
including
genomic
instability,
epigenetic
changes,
mitochondrial
dysfunction,
inflammation,
impaired
nutrient
sensing,
altered
intercellular
communication.
Propolis,
rich
array
bioactive
compounds,
functions
as
potent
functional
food,
modulating
metabolism,
gut
microbiota,
immune
response,
offering
significant
health
benefits.
Studies
emphasize
propolis'
properties,
such
antitumor,
cardioprotective,
neuroprotective
effects,
well
ability
mitigate
oxidative
stress,
DNA
damage,
pathogenic
bacteria
growth.

Background/Objectives:
Ovarian
cancer
remains
one
of
the
most
commonly
diagnosed
malignancies
among
women
worldwide.
The
heterogeneity
tumor
subtypes
and
emergence
treatment
resistance
have
raised
significant
concerns
regarding
long-term
efficacy
chemotherapy,
radiotherapy,
immunotherapy.
In
response
to
these
challenges,
drug
repurposing
strategies—utilizing
existing
drugs
in
novel
therapeutic
contexts—have
gained
increasing
attention.
This
study
aimed
investigate
cytotoxic
apoptotic
effects
combined
application
doxorubicin
(DX)
thymoquinone
(TQ)
on
ovarian
adenocarcinoma
cells
(OVCAR3).
Methods:
OVCAR3
were
cultured
RPMI
medium
supplemented
with
10%
fetal
bovine
serum
(FBS)
1%
penicillin/streptomycin.
Cell
viability
proliferation
assessed
using
MTT
assay
following
various
concentrations
DX
TQ.
NucBlue
immunofluorescence
staining
was
employed
examine
nuclear
morphology
identify
apoptosis-associated
changes.
Additionally,
quantitative
real-time
polymerase
chain
reaction
(qRT-PCR)
per-formed
evaluate
expression
levels
apoptosis-related
oncogenic
pathway
genes,
including
RAF,
RAS,
Bcl-2,
Bax.
Results:
results
demonstrated
that
combination
TQ
significantly
reduced
cell
induced
apoptosis
a
dose-dependent
manner.
qRT-PCR
analysis
revealed
downregulation
Bcl-2
expression,
along
an
upregulation
Bax,
indicating
activation
intrinsic
pathway.
These
findings
suggest
exerts
an-ti-proliferative
pro-apoptotic
by
modulating
RAS/RAF
signaling
cascade.
Furthermore,
co-administration
potentiated
effects,
suggesting
synergistic
interaction
between
two
agents.
Conclusions:
Histopathological
molecular
evaluations
further
confirmed
suppression
key
pathways.
Collectively,
underscore
potential
as
both
monotherapy
adjuvant
conventional
warranting
validation
preclinical
clinical
studies.

Aim:
Ovarian
cancer
is
one
of
the

frequently
diagnosed
cancers
in
women
world.
Recognition

tumors

resistance,
laboratories
implemented
diversion
system
by
putting
old
drugs
into
new
forms.

this
study,
cytotoxic
apoptotic
effects
combination
doxorubicin

thymoquinone
(TQ)
on
ovarian
adenocarcinoma
cells
(OVCAR3)
were
investigated.
Material
Method:
OVCAR3
cell
line
was
cultured
RPMI
