A bispecific thermosensitive system was created to deliver drugs and aid T cells in identifying cancer for enhancing the tumor-killing effect. Androgenic alopecia (AGA) is a prevalent progressive hair loss condition. The main therapeutic drug, minoxidil, limited by its poor efficacy and side effects such as contact dermatitis hypertrichosis. Nitric oxide (NO), an endothelial-derived relaxing factor, promotes angiogenesis accelerates blood flow, enhancing nutrient supply similar to minoxidil. Accordingly, we utilized poly(vinyl alcohol) film (PVA) loaded with hyaluronic acid (HA) liposomes construct multistage transdermal NO delivery system (PVA@HL/NONOate) for the treatment of AGA. HA provided efficient loading extended release, while PVA improved skin penetration sustained increasing bioavailability. Low-concentration effectively enhanced follicle vitality repaired vessels. Mechanistically, low-concentration could treat AGA mainly regulating HIF-1 signaling pathway promote angiogenesis, reducing inflammation downregulating expression TNFRSF9 IL-6, repairing follicles genes in CXCL5-IL-17 inflammatory axis.
