Drosophila suzukii severely damages the production of berry and stone fruits in large parts world. Unlike D. melanogaster, which reproduces on overripe fermenting ground, prefers to lay its eggs ripening still plants. Flies locate fruit hosts by their odorant volatiles, are detected encoded a highly specialised olfactory system before being translated into behaviour. The exact information-processing pathway is not yet fully understood, especially evaluation odour attractiveness. It also unclear what differentiates brains melanogaster cause crucial difference host selection. We hypothesised that basis for different behaviours already formed at level antennal lobe neuronal responses volatiles associated with ripe fruit. thus investigated 3D vivo two-photon calcium imaging how both species odours from fruits, leaves, fermented bacteria, mixtures lobe. then assessed behavioural odours. neural reflect species-dependent shifts code. In addition this, morphological differences were observed. However, this was directly reflected tested.

Recent advances in calcium imaging, including the development of fast and sensitive genetically encoded indicators, high-resolution camera chips for wide-field resonant scanning mirrors laser microscopy, have notably improved temporal spatial resolution functional imaging analysis. Nonetheless, variability approaches brain structures challenges versatile reliable segmentation methods. Standard techniques, such as manual selection regions interest or machine learning solutions, often fall short due to either user bias, non-transferability among systems, computational demand. To overcome these issues, we developed CalciSeg, a data-driven reproducible approach unsupervised data segmentation. CalciSeg addresses associated with structure bias by offering computationally efficient solution automatic image based on two parameters: regions' size limits number refinement iterations. We evaluated efficacy datasets varied complexity, different insect species (locusts, bees, cockroaches), systems (wide-field, confocal, multiphoton), showing robustness generality our approach. Finally, user-friendly nature open-source availability facilitate integration this algorithm into existing analysis pipelines.

Abstract Neonicotinoids represent over a quarter of the global pesticide market. Research on their environmental impact has revealed adverse effect cognitive functions pollinators, in particular bees. Cognitive impairments, mostly by behavioural studies, are phenotypic expression an alteration underlying neural circuits, matter deserving greater attention. Here, we reviewed studies field‐relevant doses neonicotinoids neurophysiology and neurodevelopment In particular, focus olfactory system as much knowledge been gained different brain areas that participate odour processing. Recent have detrimental effects at multiple levels system, including modulation odorant‐induced activity sensory neurons, diminished responses antennal lobe (the first processing centre) abnormal development connectivity within mushroom bodies (central neuropils involved multisensory integration, learning memory storage, among others). Given importance perception for aspects bee biology, reported disruption circuit, which can occur even upon exposure to sublethal neonicotinoids, severe consequences both individual colony levels. Moreover, multimodal structure such indicate neonicotinoids' translates other domains. Assessing pesticides is crucial understanding how influence behaviour ecological contexts defining effective sustainable agricultural practices.
