Complex diseases may not always progress in a gradual manner. In the early stages of complex diseases, obvious symptoms are usually observable, but there is commonality: brief state predisease between progression from normal to disease state, which includes three stages: critical predisease, and state. Identifying this especially with single sample an individual, remains difficult task. study, we applied methods, i.e., single-sample Jensen–Shannon Divergence (sJSD), network information gain (NIG), temporal flow entropy (TNFE) method, simulated dataset type 2 diabetes (GSE13268 GSE13269). Three different methods were utilized create indexes, including Inconsistency Index (ICI), NIG, TNFE, measure overall disruption caused by individual samples compared set reference samples. Abstract The rapid evolution of high-throughput sequencing technologies has revolutionized biomedical research, producing vast amounts gene expression data that hold immense potential for biological discovery and clinical applications. Effectively mining these large-scale, high-dimensional is crucial facilitating disease detection, subtype differentiation, understanding the molecular mechanisms underlying progression. However, conventional paradigm single-gene profiling, measuring absolute levels individual genes, faces critical limitations in implementation. These include vulnerability to batch effects platform-dependent normalization requirements. In contrast, emerging approaches analyzing relative relationships between pairs demonstrate unique advantages. By focusing on binary comparisons two genes’ magnitudes, methods inherently normalize experimental variations while capturing biologically stable interaction patterns. this review, we systematically evaluate pair-based analytical frameworks. We classify eleven computational into fundamental categories: value-based quantifying differential patterns, rank-based exploiting transcriptional ordering relationships. To bridge methodological development with practical implementation, establish a reproducible pipeline incorporating feature selection, classifier construction, model evaluation modules using real-world benchmark datasets from pulmonary tuberculosis studies. findings position pair analysis as transformative omics data, direct implications precision biomarker mechanistic studies

Myocardial hypertrophy is a chronic cardiac condition that often occurs from long-term pressure or volumetric load on the heart. Propranolol hydrochloride has been employed in research hypertension, pheochromocytoma, myocardial infarction, arrhythmias, angina pectoris, and hypertrophic cardiomyopathy. Current treatments for this have side effects, such as arrhythmias cell death, thus necessitating safer more effective alternatives. Recently, natural products gained attention drug development because of their low toxicity high efficacy. Cardamonin, compound derived Chinese herbal materials, shown potential inhibiting oxidative stress inflammation, which beneficial cardiovascular health. Nevertheless, impact remodeling still uncertain METHODS: Approach We transverse aortic constriction (TAC)model to simulate pathological conditions hypertrophy. Mice were administered varying doses CAR (10 40 mg kg
