In rheumatoid arthritis, dysregulated cytokine signaling has been implicated as a primary factor in chronic inflammation. Many antirheumatic and biological therapies are used to suppress joint inflammation, but despite these advances, effectiveness is not universal, delivery often at high doses, which can predispose patients significant off-target effects. During the inappropriate regulation of factors by macrophages accelerates progression disease driving an imbalance inflammatory cytokines, making ideal cellular target. To develop macrophage-based therapy treat we engineered novel induced pluripotent stem cell (iPSC)-derived macrophage capable delivering soluble TNF receptor 1 (TNFR1), anti-inflammatory biologic inhibitor tumor necrosis alpha (TNF-α), auto-regulated manner response TNF-α. Ulcerative colitis (UC) is a significant inflammatory bowel disease caused by an abnormal immune response to gut microbes. However, there are still gaps in our understanding of how and metabolic changes specifically contribute this disease.
