Neoadjuvant chemotherapy (NAC) for rectal cancer (RC) shows promising clinical response. The modulation of the tumor microenvironment (TME) by NAC and its association with therapeutic response remain unclear. Here, we use single-cell RNA sequencing spatial transcriptome to examine cell dynamics in 29 patients RC, who are sampled pairwise before after treatment. We construct a high-resolution cellular dynamic landscape remodeled their associations markedly reshapes populations cancer-associated fibroblasts (CAFs), which is strongly associated CAF subsets regulate TME through recruitment crosstalk activate immunity suppress progression multiple cytokines, including CXCL12, SLIT2, DCN. In contrast, epithelial-mesenchymal transition malignant cells upregulated CAF_FAP MIR4435-2HG induction, resulting worse outcomes. Our study demonstrates that inhibits modulates remodeling CAFs.

Background Obesity is a metabolic and chronic inflammatory disease involving genetic environmental factors. This study aimed to investigate the causal relationship among gut microbiota abundance, plasma metabolomics, peripheral cell (blood immune cell) counts, cytokines, obesity. Methods Summary statistics of 191 traits (N = 18,340), 1,400 metabolite 8,299), 128 counts cells, N 408,112; 3,757), 41 cytokine 8,293), 6 obesity were obtained from publicly available genome-wide association studies. Two-sample Mendelian randomization (MR) analysis was applied infer links using inverse variance-weighted, maximum likelihood, MR-Egger, weighted median, mode, Wald ratio methods. Several sensitivity analyses also utilized ensure reliable MR results. Finally, we used mediation identify pathway mediated by metabolites, cytokines. Results revealed effect 44 taxa, 281 27 8 cytokines on Among them, five shared taxa belonged phylum Actinobacteria , order Bifidobacteriales family Bifidobacteriaceae genus Lachnospiraceae UCG008, species Eubacterium nodatum group. Furthermore, screened 42 7 1 cytokine. Based known observed that pathways D-arginine, D-ornithine, linoleic acid, glycerophospholipid metabolism closely related 20 relationships, including obesity, 17 2 Sensitivity represented no heterogeneity or pleiotropy in this study. Conclusion Our findings support microbiota, These biomarkers provide new insights into mechanisms underlying contribute its prevention, diagnosis, treatment.

Abstract This study aimed to investigate the serum and expression levels of C-X-C motif chemokine ligand 9 (CXCL9), CXCL10, CXCL11, CXC receptor 3 (CXCR3) in minor salivary glands (MSGs) patients with primary Sjögren’s syndrome (pSS), explore their correlations clinical parameters. Serum samples from 49 diagnosed pSS, 33 rheumatoid arthritis (RA), 30 healthy controls (HCs) were collected for measurements CXCL9, CXCR3. Additionally, CXCL MSG tissues measured 41 who underwent biopsy. Correlations between CXCL/CXCR serum/MSG factors/salivary scintigraphy parameters analyzed. CXCL11 CXCR3 showed statistically significant differences among pSS RA HCs (serum pSS:RA:HC = 235.6 ± 500.1 pg/mL:90.0 200.3 pg/mL:45.9 53.6 pg/mL; p 0.041, CXCR3, 3.27 1.32 ng/mL:3.29 1.17 ng/mL:2.00 1.12 ng/mL; < 0.001). CXCL10 a difference (64.5 54.2 pg/mL) (18.6 18.1 pg/mL, 0.001), while CXCL9 did not exhibit groups. Correlation analysis factors revealed that positively correlated erythrocyte sedimentation rate ( r 0.524, 0.001 0.707, 0.001, respectively), total protein 0.375, 0.008 0.535, globulin 0.539, 0.639, European Alliance Associations Rheumatology SS Disease Activity Index 0.305, 0.033 0.321, 0.025). negatively Schirmer test score − 0.354, 0.05), biopsy focus 0.612, 0.02). Cell-based therapies are used extensively in tissue engineering because of their great biosafety and diverse biological functions. Among them, macrophage lysate provides a comprehensive rich network cytokines to modulate the local immune microenvironment. However, effective strategies for lysate-based biomaterials with immune-regulatory function still lacking. In this study, we engineered silk fibroin hydrogel loaded THP-1 whole-cell regulate microenvironment promote vascularized bone regeneration diabetic defects. Specifically, by modulating expression pattern key enzyme energy metabolism (mitochondrial phosphoenolpyruvate carboxykinase, PCK2) cells, generated enriched anti-inflammatory factors. This facilitated osteogenic differentiation BMSCs angiogenesis HUVECs vitro, incorporating immunomodulatory into enhanced diabetes. Mechanistically, revealed that PCK2 activated nuclear factor-kappa B (NF-κB) signaling pathway macrophages using proteomic profiling. research new insights design cell-derived biomaterials, aiming improve therapeutic outcomes pathological
