In Drosophila eight insulin-like peptides (DILP1-8) are encoded on separate genes. These DILPs characterized by unique spatial and temporal expression patterns during the lifecycle. Whereas functions of several have been extensively investigated at different developmental stages, role DILP8 signaling is primarily known from larvae pupae where it couples organ growth transitions. adult female flies, a study showed that specific set neurons express receptor, Lgr3, involved in regulation reproductive behavior. Here, we further dilp8/DILP8 Lgr3 flies functional signaling. The only site found both dilp8 immunolabeling was follicle cells mature ovaries. detected numerous brain ventral nerve cord, small peripheral innervating abdominal heart, as well close to oviduct. Ovulation affected mutants after dilp8-RNAi using cell Gal4 drivers. More eggs were retained ovaries fewer laid, indicating important for ovulation. Our data suggest signals locally expressing systemically efferent ganglia innervate oviduct muscle. Thus, may act two targets regulate ovulation: rupture contractions. Furthermore, could show manipulations affect food intake starvation resistance. Possibly this reflects feedback between CNS ensures nutrients ovary development. summary, seems reproduction an ancient function, conserved relaxin mammals.

Environmental factors challenge the physiological homeostasis in animals, thereby evoking stress responses. Various mechanisms have evolved to counter at organism level, including regulation by neuropeptides. In recent years, much progress has been made on and neuropeptides that regulate responses metabolic/nutritional stress, as well those involved countering osmotic ionic stresses. Here, we identified a peptidergic pathway links these types of regulatory functions. We uncover neuropeptide Corazonin (Crz), previously implicated metabolic neuroendocrine factor inhibits release diuretic hormone, CAPA, modulates tolerance stress. Both knockdown Crz acute injections peptide impact desiccation recovery from chill-coma. Mapping receptor ( CrzR ) expression three pairs Capa- expressing neurons (Va neurons) ventral nerve cord mediate effects Crz. show acts restore water/ion inhibiting CAPA via inhibition cAMP production Va neurons. Knockdown affects signaling, consequently increases for desiccation, starvation, but delays chill-coma recovery. Optogenetic activation stimulates excretion simultaneous CAPA-expressing reduces this response, supporting inhibitory action Thus, maintain homeostasis, which turn tolerance. Earlier work demonstrated systemic signaling restores nutrient levels promoting food search feeding. Here additionally propose also ensures suppressing diuresis. ameliorates stress-associated physiology through modulation both neurosecretory cells fat body Drosophila .

Diapause is a physiological adaptation to conditions that are unfavorable for growth or reproduction. During diapause, animals become long-lived, stress-resistant, developmentally static, and non-reproductive, in the case of diapausing adults. has been observed at all developmental stages both vertebrates invertebrates. In adults, diapause traits weaken into adaptations such as hibernation, estivation, dormancy, torpor, which represent evolutionarily diverse versions traditional traits. These regulated through modifications endocrine program guiding development. insects, this typically includes changes molting hormones, well metabolic signals limit while skewing organism’s energetic demands toward conservation. While much work done characterize these modifications, interactions between hormones their downstream consequences incompletely understood. The current state endocrinology reviewed here highlight relevance beyond its use model study seasonality Specifically, insect an emerging mechanisms determine lifespan. induction represents dramatic change normal progression age. Hormones juvenile hormone, 20-hydroxyecdysone, prothoracicotropic hormone well-known modulate plasticity. diapause—and by extension, cessation aging—is coordinated pathways. However, research directly connecting biology aging lacking. This review explores connections perspective signaling. fields suggests appreciable overlap will greatly contribute our understanding lifespan determination.

Propagation of intercellular calcium waves through tissues has been found to coordinate different multicellular responses. Nevertheless, our understanding how operate remains limited. In this study, we explore the real-time dynamics in Drosophila adipose tissues. We identify Adipokinetic Hormone (AKH), fly functional homolog glucagon, as key factor driving Ca2+ activities tissue. find that AKH, which is released into hemolymph from AKH-producing neurosecretory cells, stimulates larval fat by a previously unrecognized gap-junction-independent mechanism promote lipolysis. adult body, however, gap-junction-dependent are triggered presumably uniformly diffused AKH. Additionally, discover amino acids activate leading increased intracellular and AKH secretion. Altogether, show dietary regulate release cells brain, subsequently tissue, enhancing lipid metabolism. Sex differences in whole-body fat storage exist many species. For example, Drosophila females store more than males. Yet, the mechanisms underlying this sex difference remain incompletely understood. Here, we identify a key role for determination gene transformer ( tra ) regulating male-female storage. Normally, functional Tra protein is present only females, where it promotes female sexual development. We show that loss of reduced storage, whereas gain males augmented Tra’s promoting was largely due to its function neurons, specifically Adipokinetic hormone (Akh)-producing cells (APCs). Our analysis Akh pathway regulation revealed male bias APC activity and function, sex-biased influenced by limiting triglyceride accumulation Importantly, increased activity, genetically manipulating rescued Tra-dependent effects on This identifies sex-specific as one mechanism levels, provides important insight into conserved dimorphism

Abstract Severe sleep deprivation (SD) has been highly associated with systemic energy wasting, such as lipid loss and glycogen depletion. Despite immune dysregulation neurotoxicity observed in SD animals, whether how the gut-secreted hormones participate SD-induced disruption of homeostasis remains largely unknown. Using Drosophila a conserved model organism, we characterize that production intestinal Allatostatin A (AstA), major gut-peptide hormone, is robustly increased adult flies bearing severe SD. Interestingly, removal AstA gut using specific drivers significantly improves depletion without affecting homeostasis. We reveal molecular mechanisms whereby promotes release an adipokinetic hormone (Akh), insulin counter-regulatory functionally equivalent to mammalian glucagon, mobilize reserves by remotely targeting its receptor AstA-R2 Akh-producing cells. Similar regulation glucagon secretion wasting AstA/galanin also mice. Further, integrating single-cell RNA sequencing genetic validation, uncover results ROS accumulation augment via TrpA1. Altogether, our demonstrate essential roles mediating SD-associated wasting.
