The lacrimal gland (LG) secretes aqueous tears. Previous studies have provided insights into the cell lineage relationships during tissue morphogenesis. However, little is known about types composing adult LG and their progenitors. Using scRNAseq, we established first comprehensive atlas of mouse to investigate hierarchy, its secretory repertoire, sex differences. Our analysis uncovered complexity stromal landscape. Purpose: Chronic jet lag (CJL) is known to disrupt circadian rhythms, which regulate various physiological processes, including ocular surface homeostasis. However, the specific effects of CJL on lacrimal gland function and underlying cellular mechanisms remain poorly understood. Methods: A model was established using C57BL/6J mice. Extraorbital glands (ELGs) were collected at 3-hour intervals for RNA extraction high-throughput sequencing. Circadian transcriptomic profiles analyzed, functional annotations performed. Hydrogen peroxide levels total antioxidant capacity in tear fluid measured chemometric assays. Immunofluorescence used assess cell proliferation, apoptosis, immune infiltration ELGs, reactive oxygen species (ROS) accumulation. The potential therapeutic alpha-lipoic acid (ALA) CJL-induced oxidative stress pathological changes ELGs also investigated. Sleep loss markedly affects the structure and function of lacrimal gland may cause ocular surface disease as a common public health problem. This study aims to investigate circadian disturbance caused by sleep leading dysfunction extraorbital glands (ELGs).A mouse deprivation (SD) model for studies was built in C57BL/6J male mice. After four weeks, ELGs were collected at three-hour intervals during 24-hour period. The Jonckheere-Terpstra-Kendall algorithm used determine composition, phase, rhythmicity transcriptomic profiles ELGs. Furthermore, we compared non-sleep-deprived SD-treated ELG (i) reactive oxygen species (ROS) fluorescein staining, (ii) DNA damage immunostaining γ-H2Ax, (iii) migration immune cells CD4, CD8, γδ-TCR, CD64, CX3CR1. Objectives: To evaluate the effect of astaxanthin in treatment mild-to-moderate dry eye disease (DED) middle-aged and elderly patients. Methods: 120 eyes 60 patients with DED were enrolled this prospective, one-group, quasi-experimental study. Six milligram Astaxanthin tablets (Weihong Haematococcus Pluvialis Astaxanthin, Hangzhou Xinwei Low Carbon Technology R&D Co., Ltd., China) administered orally, twice daily for 30 ± 2 days. History diseases, treatment, systemic disease, medication before test recorded. In addition, ocular surface index (OSDI) questionnaire, non-invasive tear break-up time (NIBUT), fluorescein (FBUT), corneal staining (CFS) score, eyelid margin signs, meibomian gland (MG) expressibility, meibum quality, dropout (MGDR), Schirmer I (SIt), meniscus height (TMH), bulbar conjunctiva congestion degree, blink frequency, incomplete rate, thickness film lipid layer collected weeks after initiation at end treatment. Visual acuity (VA), intraocular pressure (IOP), anterior segment, fundus, discomfort symptoms other adverse reactions also monitored throughout study to assess safety. Results: OSDI NIBUT, BUT, CFS MG frequency improved significantly varying degrees compared those (P < 0.05), while TMH, SIt, conjunctival congestion, layer, MGDR, VA IOP did not differ > 0.05). Conclusions: Oral administration improves signs DED.
