Abstract Extracellular vesicles (EVs) have been shown to be involved in cell‐cell communication and take part both physiological pathological processes. Thanks their exclusive cargo, which includes proteins, lipids, nucleic acids from the originating cells, they are gaining interest as potential biomarkers of disease. In recent years, appealing features fascinating researchers all over world, thus increasing number vitro studies focused on EV release, content, biological activities. Cultured cell lines most‐used source EVs; however, EVs released cultures influenced by culture conditions, such use foetal bovine serum (FBS). FBS is most common supplement for media, but it also a contaminants, exogenous EVs, RNA, protein aggregates, that can contaminate cell‐derived influence cargo composition. The presence contaminants samples well‐known issue limits clinical applications need standardization. this review, we will discuss pros cons using well protocols used remove FBS.

Abstract Background Porcine seminal plasma (SP) is endowed with a heterogeneous population of extracellular vesicles (sEVs). This study evaluated the immunophenotypic profile by high-sensitivity flow cytometry eight sEV subpopulations isolated according to their size (small [S-sEVs] and large [L-sEVs]) from four different SP sources, namely three ejaculate fractions (the first 10 mL sperm rich fraction [SRF-P1], remaining SRF [SRF-P2], post-SRF [PSRF]) entire (EE). Methods Seminal EVs were using exclusion chromatography-based protocol six pools (five ejaculates/pool) each source characterized complementary approaches including total protein (BCA™assay), particle distribution (dynamic light scattering), morphology (transmission electron microscopy), purity (albumin Western blot). Expression CD9, CD63, CD81, CD44 HSP90β was analyzed in all MIFlowCyt-EV guidelines, an accurate calibration, controls, discrimination CFSE-labelling. Porcine seminal plasma (SP) is loaded with a heterogeneous population of extracellular vesicles (sEVs) that modulate several reproductive-related processes. Seminal plasma contains many morphologically heterogeneous extracellular vesicles (sEVs). These are sequentially released by cells of the testis, epididymis, and accessory sex glands involved in male female reproductive processes. This study aimed to define depth sEV subsets isolated ultrafiltration size exclusion chromatography, decode their proteomic profiles using liquid chromatography-tandem mass spectrometry, quantify identified proteins sequential window acquisition all theoretical spectra. Seminal plasma contains numerous extracellular vesicles (sEVs). Since sEVs are apparently involved in male (in)fertility, this systematic review focused on studies specifically investigating such relationship. Embase, PubMed, and Scopus databases were searched up to 31 December 2022, primarily identifying a total of 1440 articles. After processing for screening eligibility, 305 selected as they sEVs, 42 them considered eligible because included the word fertility or related infertility, subfertility, fertilization, recurrent pregnancy loss title, objective(s), and/or keywords. Only nine met inclusion criteria, namely (a) conducting experiments aimed at associating with concerns (b) isolating adequately characterizing sEVs. Six conducted humans, two laboratory animals, one livestock.
