Abstract
Extracellular
vesicles
(EVs)
have
been
shown
to
be
involved
in
cell‐cell
communication
and
take
part
both
physiological
pathological
processes.
Thanks
their
exclusive
cargo,
which
includes
proteins,
lipids,
nucleic
acids
from
the
originating
cells,
they
are
gaining
interest
as
potential
biomarkers
of
disease.
In
recent
years,
appealing
features
fascinating
researchers
all
over
world,
thus
increasing

vitro
studies
focused
on
EV
release,
content,
biological
activities.
Cultured
cell
lines
most‐used
source
EVs;
however,
EVs
released
cultures
influenced
by
culture
conditions,

bovine
serum
(FBS).
FBS
is
most
common
supplement
for
media,
but
it
also
a
contaminants,
exogenous
EVs,
RNA,
protein
aggregates,
that
can
contaminate
cell‐derived

Abstract
Background
Porcine
seminal
plasma
(SP)
is
endowed
with
a
heterogeneous
population
of
extracellular
vesicles
(sEVs).
This
study
evaluated
the
immunophenotypic
profile
by
high-sensitivity
flow
cytometry
eight
sEV
subpopulations
isolated
according
to
their
size
(small
[S-sEVs]
and
large
[L-sEVs])
from
four
different
SP
sources,
namely
three
ejaculate
fractions
(the
first
10
mL
sperm
rich
fraction
[SRF-P1],
remaining
SRF
[SRF-P2],
post-SRF
[PSRF])
entire
(EE).
Methods
Seminal
EVs
were

exclusion
chromatography-based
protocol

pools
(five
ejaculates/pool)
