Psoriasis is a chronic and incurable skin inflammation driven by an abnormal immune response. Our study aims to investigate the potential of interferon-γ (IFN-γ) primed mesenchymal stem cells (IMSCs) in targeting T attenuate psoriasis-like inflammation, elucidate underlying molecular mechanism involved. Mesenchymal (MSCs) were isolated from umbilical cord identified based on their surface markers. models established then treated with IMSCs. Flow cytometry analysis was used examine cell markers percentages. Indoleamine-2,3-dioxygenase (IDO) knocked down small interfering RNA (siRNA) examined western blot assay. Background: Psoriasis is a chronic inflammatory skin disease. However, the influence of TOP2A and MELK genes on psoriasis remains unclear. Methods: datasets GSE166388 GSE181318 were downloaded from Gene Expression Omnibus (GEO) database generated GPL570 GPL22120. Differential gene expression (DEGs) was identified. Functional enrichment analysis, set analysis (GSEA), weighted co-expression network (WGCNA), immune infiltration conducted. The protein-protein interaction (PPI) constructed analyzed. heat map generated. most relevant diseases associated with core determined through comparison Comparative Toxicogenomics Database (CTD) website. TargetScan used to select miRNAs regulating central DEGs. Results: A total 773 DEGs According Ontology (GO) they mainly enriched in mitochondrial expression, oxidative phosphorylation, envelope, mitochondria ribosome. Kyoto Encyclopedia Genes Genomes (KEGG) revealed that target cells metabolic pathways, proteasome, phosphorylation. Seven (TOP2A, NUF2, MELK, ASPM, DLGAP5, CCNA2, DEPDC1B) obtained. heatmap showed high MELK) samples, while DEPDC1B, ASPM lowly expressed samples. CTD found related neoplasms, diseases, psoriasis, erythema, dermatitis, infections. Conclusion: are highly higher poorer prognosis.
