Turmeric (Curcuma longa) is a type of herb belonging to ginger family, which widely grown in southern and south western tropical Asia region. Turmeric, has an importance place the cuisines Iran, Malesia, India, China, Polynesia, Thailand, often used as spice effect on nature, color, taste foods. also known have been for centuries India China medical treatments illnesses such dermatologic diseases, infection, stress, depression. Turmeric's effects health are generally centered upon orange-yellow colored, lipophilic polyphenol substance called "curcumin," acquired from rhizomes herb. Curcumin recently antioxidant, anti-inflammatory, anticancer and, thanks these effects, important role prevention treatment various ranging notably cancer autoimmune, neurological, cardiovascular diabetic. Furthermore, it aimed increase biological activity physiological curcumin body by synthesizing analogues. This article reviews history, chemical physical features, analogues, metabolites, mechanisms its activities, curcumin.

In the present perspective, we give a critical review on coordination chemistry of metal ions copper and zinc to amyloid-beta (Abeta) peptide; such complexes have been linked Alzheimer's disease. We focus two main issues: identification sphere Cu(ii) Zn(ii) affinity these towards peptide. With aim come up with as few possible valuable structural models binding values, critically divergent propositions reported in literature take into account experimental differences limits methods used published studies. propose that: (i) conditional dissociation constant Cu(Abeta) lies range 10 pM 100 nM, preference for region between 1 nM. (ii) Two most likely modes predominant form at physiological pH can be retained, both being 3N1O distorted square planar. first model, ion is coordinated by Ntau atoms three His residues carboxylate Asp1. second N-terminus functions Asp1 are ligated together His6 His13 (or His14). An equilibrium forms room temperature, preferentially freezing out one would explain divergences results (in particular, those obtained EPR NMR). (iii) The apparent constants Zn(Abeta) various buffers 20 muM (a times lower estimated. (iv) For coordination, implication consensual. and/or functions. Additional ligands possible, Glu11 or H(2)O.

Alzheimer disease (AD) is an age-related neurodegenerative disease. AD characterized by progressive cognitive impairment. One of the main histopathological hallmarks brain presence senile plaques (SPs) and another elevated oxidative stress. The component SPs amyloid beta-peptide (Aβ) that derived from proteolytic cleavage precursor protein.

Neurodegenerative diseases pose a substantial socioeconomic burden on society. Unfortunately, the aging world population and lack of effective cures foreshadow negative outlook. Although large amount research has been dedicated to elucidating pathologies neurodegenerative diseases, their principal causes remain elusive. Metal ion dyshomeostasis, proteopathy, oxidative stress, neurotransmitter deficiencies are pathological features shared across multiple disorders. In addition, these factors proposed be interrelated upon disease progression. Thus, development multifunctional compounds capable simultaneously interacting with several components suggested as solution undertake complex diseases. this review, we outline discuss possible therapeutic targets in Alzheimer's disease, Parkinson's amyotrophic lateral sclerosis molecules, previously designed or discovered potential drug candidates for disorders emphasis multifunctionality. underrepresented areas discussed indicate new directions.

Reactive oxygen species (ROS) are produced at low levels in mammalian cells by various metabolic processes, such as oxidative phosphorylation the mitochondrial respiratory chain, NAD(P)H oxidases, and arachidonic acid metabolism. To maintain physiological redox balance, have endogenous antioxidant defenses regulated transcriptional level Nrf2/ARE. Oxidative stress results when ROS production exceeds cell's ability to detoxify ROS. Overproduction of damages cellular components, including lipids, leading decline function cell death. Reaction with lipids produces oxidized phospholipids, which give rise 4-hydroxynonenal, 4-oxo-2-nonenal, acrolein. The brain is susceptible damage due its high lipid content consumption. Neurodegenerative diseases (AD, ALS, bipolar disorder, epilepsy, Friedreich's ataxia, HD, MS, NBIA, NPC, PD, peroxisomal disorders, schizophrenia, Wallerian degeneration, Zellweger syndrome) CNS traumas (stroke, TBI, SCI) problems vast clinical importance. Free iron can react H(2)O(2) via Fenton reaction, a primary cause peroxidation, may be particular importance for these injuries disorders. Cholesterol an important regulator organization precursor neurosteroid biosynthesis. Atherosclerosis, major risk factor ischemic stroke, involves accumulation LDL arteries, foam formation plaque development. This review will discuss role oxidation/peroxidation injuries/disorders.
