Lateral flow assay (LFA)-based rapid antigen tests are experiencing extensive global uptake as an expeditious and highly effective modality for the screening of viral infections during COVID-19 pandemic. While these devices have played a significant role in alleviating burden on public healthcare system, their specificity sensitivity fall short compared with molecular tests. In this study, we endeavor to address both limitations through utilization DNA nanotechnology LFA format, wherein substitute target-specific antibody designer nanostructure-based probes recognizing SARS-CoV-2 virus via multivalent, pattern-matching interactions. We meticulously designed Net-shaped nanostructure strategically arranged trimeric clusters aptamers that specifically recognize spike proteins SARS-CoV-2. This approach has proven instrumental bolstering virus-binding affinity LFAs. Our findings indicate high sensitivity, enabling detection loads ranging from 103 108 copies/mL. notable is maintained across various strains, obviating need intricate sample preparation protocols. The significance heightened lies crucial by nanostructure, which facilitates extremely low levels loads. not only enhances overall reliability self-testing but also reduces likelihood false-negative results, especially cases load within patient samples.

Broad-spectrum antiviral platforms are always desired but still lack the ability to cope with threats global public health. Herein, we develop a poly aptamer encoded DNA nanocatcher platform that can trap entire virus particles inhibit infection broad spectrum. Ultralong single-stranded (ssDNA) containing repeated aptamers was synthesized as scaffold of via biocatalytic process, wherein mineralization magnesium pyrophosphate on ssDNA could occur and consequently lead formation interfacial nanocaves decorated virus-binding aptamers. Once viruses were recognized by apatmers, they would be captured trapped in multisite synergistic interactions. Meanwhile, size nanocatchers optimized prevent their cellular uptake, which further guaranteed inhibition infection. This review provides a comprehensive summary of evidence to explore the role and value differential diagnosis in management Acute Respiratory Infections (ARIs) through point-of-care (POC) rapid testing post-pandemic scenario, paying particular attention coronavirus disease 2019 (COVID-19), influenza, respiratory syncytial virus (RSV).
