Abstract Aging and cancer exhibit apparent links that we will examine in this review. The null hypothesis aging coincide because both are driven by time, irrespective of the precise causes, can be confronted with idea share common mechanistic grounds referred to as ‘hallmarks’. Indeed, several hallmarks also contribute carcinogenesis tumor progression, but some molecular cellular characteristics may reduce probability developing lethal cancer, perhaps explaining why very old age (> 90 years) is accompanied a reduced incidence neoplastic diseases. We discuss possibility process itself causes meaning time-dependent degradation supracellular functions accompanies produces byproduct or ‘age-associated disease’. Conversely, its treatment erode health drive process, has dramatically been documented for survivors diagnosed during childhood, adolescence, young adulthood. conclude connected superior including endogenous lifestyle factors, well bidirectional crosstalk, together render not only risk factor an important parameter must considered therapeutic decisions.

Abstract The accumulation of senescent cells within tissues is a hallmark the aging process. Senescent are also commonly present in many age-related diseases and cancer microenvironment. escape abnormal from immune surveillance indicates that there some defect function cytotoxic cells, e.g., CD8 + T natural killer (NK) cells. Recent studies have revealed expression programmed death-ligand 1 (PD-L1) protein abundantly increased An increase amount PD-L1 protects clearance by PD-1 checkpoint receptor In fact, activation suppresses properties NK promoting state immunosenescence. inhibitory PD-1/PD-L1 pathway acts cooperation with immunosuppressive cells; for example, can enhance differentiation regulatory (Treg), myeloid-derived suppressor (MDSC), M2 macrophages, whereas cytokines secreted stimulate protein. Interestingly, signaling pathways known to promote cellular senescence process crucial stimulators protein, epigenetic regulation, inflammatory mediators, mTOR-related signaling, cGAS-STING pathway, AhR signaling. It seems axis has role thus it promotes tissues. Thus, blockade might be potential anti-aging senolytic therapy. Redox signaling acts as a critical mediator in the dynamic interactions between organisms and their external environment, profoundly influencing both onset progression of various diseases. Under physiological conditions, oxidative free radicals generated by mitochondrial respiratory chain, endoplasmic reticulum, NADPH oxidases can be effectively neutralized NRF2-mediated antioxidant responses. These responses elevate synthesis superoxide dismutase (SOD), catalase, well key molecules like nicotinamide adenine dinucleotide phosphate (NADPH) glutathione (GSH), thereby maintaining cellular redox homeostasis. Disruption this finely tuned equilibrium is closely linked to pathogenesis wide range Recent advances have broadened our understanding molecular mechanisms underpinning dysregulation, highlighting pivotal roles genomic instability, epigenetic modifications, protein degradation, metabolic reprogramming. findings provide foundation for exploring regulation mechanistic basis improving therapeutic strategies. While antioxidant-based therapies shown early promise conditions where stress plays primary pathological role, efficacy diseases characterized complex, multifactorial etiologies remains controversial. A deeper, context-specific signaling, particularly redox-sensitive proteins, designing targeted aimed at re-establishing balance. Emerging small molecule inhibitors that target specific cysteine residues proteins demonstrated promising preclinical outcomes, setting stage forthcoming clinical trials. Aging is a natural and irreversible process, affecting living organisms by negatively impacting the tissues' cells' morphology functionality consequently being responsible for aging-related diseases. Taking into account actual preoccupations of both consumers researchers, healthy anti-aging alternatives are intensively studied in order to address such concerns. Due their functional features, plant flavonoids can be considered valuable nutraceuticals. This paper highlights possibilities use extracted from various plants potential on skin, brain, heart. Moreover, anticarcinogenic, anti-inflammatory, anti-diabetic properties summarized, along with senescence-associated mechanisms. Both nutraceutical cosmeceutical fields continuously developing originating promising candidates obtain products. Thus, bioactive compounds' extraction subsequent involvement innovative product manufacturing must carefully performed while aware intrinsic extrinsic factors that may affect phytochemicals' structures, bioavailability, health effects.

Abstract Ginkgo biloba extract is reported to have therapeutic effects on aging‐related disorders. However, the specific component responsible for this biological function and its mechanism of action remain largely unknown. This study finds that Ginkgetin, an active ingredient extract, can alleviate cellular senescence improve pathologies in multiple tissues aging mice. To reveal molecular Ginkgetin's anti‐aging effect, a graph convolutional network‐based drug “on‐target” pathway prediction algorithm employed. The results indicate cGAS‐STING may be potential target Ginkgetin. Subsequent cell biophysical data confirmed Ginkgetin directly binds carboxy‐terminal domain STING protein, thereby inhibiting activation signal transduction. Furthermore, vivo pharmacodynamic showed effectively alleviates systemic inflammation Trex1 −/− mice inhibits abnormally activated signaling mouse model. In summary, study, utilizing artificial intelligence combined with pharmacological methods, confirms serves as critical alleviating senescence. Importantly, elucidates underlying effect providing robust theoretical basis use.

ABSTRACT Longevity individuals have lower susceptibility to chronic hypoxia, inflammation, oxidative stress, and aging‐related diseases. It has long been speculated that “rejuvenation molecules” exist in their blood promote extended lifespan. We unexpectedly discovered longevity exhibit erythrocyte oxygen release function similar young individuals, whereas most elderly show reduced capacity. Untargeted metabolomics profiling revealed are characterized by youth‐like metabolic reprogramming these metabolites effectively differentiate the from elderly. Quantification analyses led us identify multiple novel longevity‐related within erythrocytes including adenosine, sphingosine‐1‐phosphate (S1P), glutathione (GSH) related amino acids. Mechanistically, we increased bisphosphoglycerate mutase (BPGM) MFSD2B protein levels of collaboratively work together induce elevation intracellular S1P, glyceraldehyde‐3‐phosphate dehydrogenase (GAPDH) membrane cytosol, thereby orchestrate glucose toward Rapoport–Luebering Shunt 2,3‐BPG production trigger delivery. Furthermore, glutamine glutamate transporter expression coupled with enhanced metabolism underlie elevated GSH higher anti‐oxidative stress capacity individuals. As such, displayed less systemic hypoxia‐related more antioxidative anti‐inflammatory plasma, healthier clinical outcomes inflammation parameters as well better glucose–lipid metabolism, liver kidney function. Overall, identified youthful enable counteract peripheral tissue thus maintaining healthspan.

Age-related diseases have emerged as a global concern the population ages. Consequently, understanding underlying causes of aging and exploring potential anti-aging interventions is imperative. In females, ovaries serve principal organs responsible for ovulation production female hormones. The are related to infertility, menopause, associated menopausal syndromes, with menopause representing culmination ovarian aging. Current evidence indicates that may contribute dysfunction across multiple organ systems, including, but not limited to, cognitive impairment, osteoporosis, cardiovascular disease. Nevertheless, due widespread distribution sex hormone receptors throughout body, affects only these specific also influences broader spectrum age-related in women. Despite this, impact on overall has been largely neglected. This review provides thorough summary diseases, encompassing nervous, circulatory, locomotor, urinary, digestive, respiratory, endocrine systems. Additionally, we outlined prospective therapeutic approaches addressing both aim mitigating their impacts preserving women's fertility, physical health, psychological well-being.
