Abstract Colorectal cancer (CRC) development and progression, one of the most common cancers globally, is supported by specific mechanisms to escape cell death despite chemotherapy, including cellular autophagy. Autophagy an evolutionarily highly conserved degradation pathway involved in a variety processes, such as maintenance homeostasis clearance foreign bodies, its imbalance associated with many diseases. However, role autophagy CRC progression remains controversial, it has dual function, affecting either or survival, senescence tumor therapy. Indeed, numerous data have been presented that serves alternative apoptosis when latter ineffective apoptosis‐resistant cells, which why also referred programmed type II. Curcumin, active constituents Curcuma longa , great potential combat influencing various signaling pathways epigenetic regulation safe cost‐effective approach. This review discusses efficacy curcumin against vitro vivo, particularly modulation pathways. While clinical studies assessed prevention treatment, none specifically examined Nonetheless, we offer overview correlations support use this polyphenol prophylactic co‐therapeutic agent CRC.

Heat shock proteins (Hsps) are involved in one of the adaptive mechanisms protecting cells against environmental and metabolic stress. Moreover, large role these carcinogenesis process, as well chemoresistance, was noticed. This review aims to draw attention possibilities using Hsps developing new cancer therapy methods, indicate directions for future research on this topic. In order discuss matter, a thorough latest scientific literature carried out, taking into account importance selected from Hsp family, including Hsp27, Hsp40, Hsp60, Hsp70, Hsp90 Hsp110. One more characteristic features all is that they play multifaceted progression, which makes them an obvious target modern anticancer therapy. Some researchers emphasize directly inhibiting action proteins. turn, others point their possible use design vaccines, would work by inducing immune response various types cancer. Due possibilities, it believed may contribute progress oncoimmunology, thus help development therapies, be characterized higher effectiveness lower toxicity patients.

The pathophysiology of osteoarthritis (OA) is closely linked to autophagy abnormalities in articular chondrocytes, the sole mature cell type healthy cartilage. Nevertheless, precise molecular mechanism remains uncertain. Previous research has demonstrated that leptin activates mTORC1 , thereby inhibiting chondrocyte during progression OA. In this study, it presence induces a substantial increase expression STAT3, leading notable decrease REDD1 and subsequent phosphorylation p70S6K, recognized downstream effector mTORC1. Conversely, inhibition yields contrasting effects. Additionally, potential advantages utilizing sustained intra-articular release inhibitor (LI) via an injectable, thermosensitive poly(D,L-lactide)-poly(ethylene glycol)-poly(D,L-lactide) (PDLLA-PEG-PDLLA: PLEL) hydrogel delivery system for purpose investigating its impact on cartilage repair are explored. study conducted LI-loaded PLEL (PLEL@LI) demonstrates remarkable efficacy OA displays encouraging therapeutic restoration subchondral bone These findings establish solid foundation advancement pioneering treatment approach PLEL@LI

Colorectal cancer (CRC) remains one of the most commonly diagnosed and deadliest types worldwide. CRC displays a desmoplastic reaction (DR) that has been inversely associated with poor prognosis; less DR is better prognosis. This generates excessive connective tissue, in which cancer-associated fibroblasts (CAFs) are critical cells form part tumor microenvironment. CAFs directly involved tumorigenesis through different mechanisms. However, their role immunosuppression not well understood, precise signal transducers activators transcription (STATs) mediating CAF activity unclear. Among myriad chemical biological factors affect CAFs, cytokines mediate function by activating STAT signaling pathways. Thus, harmful effects favoring growth invasion may be modulated using inhibitors. Here, we analyze impact STATs on immunoregulatory role.
