Abstract In recent years, heightened interest surrounds the exploration of natural phenols as potential agents for cancer therapy, specifically by inducing ferroptosis, a unique form regulated cell death characterized iron‐dependent lipid peroxidation. This review delves into roles key phenols, flavonoids, phenolic acids, curcumin, and stilbenes, in modulating ferroptosis their underlying mechanisms. Emphasizing significance amino acid, lipid, iron metabolism, study elucidates diverse pathways through which these regulate ferroptosis. Notably, well‐known polyphenol, exhibits multifaceted interactions with cellular components involved regulation, providing distinctive therapeutic avenue. Stilbenes, another class, demonstrate promising influencing metabolism processes, contributing to ferroptotic death. Understanding intricate interplay between not only illuminates complex regulatory networks but also unveils avenues novel therapies. Exploring compounds inducers presents strategy targeted treatment, capitalizing on delicate balance article synthesizes current knowledge, aiming stimulate further research context ferroptosis‐mediated therapy.

Ferroptosis is a recently discovered type of programmed cell death that mechanistically different from other types such as apoptosis, necroptosis, and autophagy. It characterized by the accumulation intracellular iron, overproduction reactive oxygen species, depletion glutathione, extensive lipid peroxidation lipids in membrane. was ferroptosis interconnected with many diseases, neurodegenerative ischemia/reperfusion injury, cancer, chronic kidney disease. Polyphenols, plant secondary metabolites known for bioactivities, are being extensively researched context their influence on which resulted great number publications showing need systematic review. In this review, an literature search performed. Databases (Scopus, Web Science, PubMed, ScienceDirect, Springer) were searched time span 2017 to November 2023, using keyword “ferroptosis” alone combination “flavonoid”, “phenolic acid”, “stilbene”, “coumarin”, “anthraquinone”, “chalcone”; after selection studies, we had 311 papers 143 phenolic compounds. total, 53 compounds showed ability induce ferroptosis, 110 able inhibit out those compounds, 20 both abilities depending model system. The most shikonin, curcumin, quercetin, resveratrol, baicalin. common modes action modulation Nrf2/GPX4 Nrf2/HO-1 axis iron metabolism.

Objectives This study employs bibliometric analysis to track evolution and identify trends of key topics in ferroptosis within the context lung cancer. By identifying emerging research areas, our aim is provide valuable insights directions for researchers this field. Methods Relevant papers reviews on cancer were retrieved from Web Science Core Collection database 5 February 2024. Bibliometric was conducted using CiteSpace 6.2.R3, VOSviewer 1.6.20, R 4.3.0, Microsoft Excel 2019. Results From 2015 2020, publications related sparse but showed a steady increase. Post-2020, there has been significant surge, with 6.4-fold increase observed by 2023. Overall, authors 4,033 institutions across 42 countries/regions contributed 606 published 262 academic journals. China emerged as leading contributor, while United States maintained dominance. Lifang Ma most prolific author, DIXON SJ, YANG WS, STOCKWELL BR being frequently co-cited. Effective communication collaboration among scholars are lacking. Key journals include Frontiers Pharmacology publication output, Nature Cell citation frequency. Research focuses molecular mechanisms ferroptosis, including endoplasmic reticulum stress, tumor microenvironment, autophagy. Therapeutic targets like GPX4, SLC7A11, P53, FSP1, Nrf2, LSH, STYK1/NOK, ACSL4 prominent. Traditional Chinese medicine also shows clinical value research. Conclusion Ferroptosis, promising avenue applications cancer, continues undergo rapid development. The iron death will remain critical focus future.

Bee pollen (BP) is one of the richest known natural resources micronutrients and bioactive phytochemicals. Some captivating bioactivities BP compounds, although being largely investigated for latter as individual molecules, remain very scarcely or completely uninvestigated in bee a whole product. Among most intriguing these bioactivities, we identified ferroptosis major one. Ferroptosis, recently discovered form cell death (connecting oxidative stress inflammation), complex pathophysiological process crucial perplexing events current challenging human diseases such cancer, neurodegeneration, general aging diseases. Many compounds were found to intricately modulate depending on cellular context by inducing this mechanism malignant cells preventing it non-malignant cells. Since research both fields, i.e., ferroptosis, still recent, deemed necessary undertake review figure out extent potential modulating mechanisms. Our proved that wide range (polyphenols, phenolamides, carotenoids, vitamins, minerals, others) substantially diverse Accordingly, phytochemicals nutrients showed interesting preclinical studies lead ferroptosis-mediated outcomes important processes, including many aging-related disorders. One paramount challenges be resolved determine how different act biological contexts, either through synergistic antagonistic behaviors. We hope our work constitutes valuable incentive future investigations promising relevant avenue.

Luteolin, a natural flavonoid compound, has demonstrated anti-inflammatory, antioxidant, and broad anti-tumor properties. Recent studies suggest that its effects are linked to enhanced CTL function-including proliferation, survival, cytotoxicity-via inhibition of the YAP/Wnt signaling pathway in tumor cells. Consequently, luteolin potential as an adjuvant combination therapies with adoptive immunotherapy. This study first assessed luteolin's tumor-inhibitory vitro vivo using cytotoxicity assays, Transwell invasion tests, wound healing analyses post-treatment growth survival time. Additionally, we explored whether combined DC/tumor fusion vaccine could synergistically enhance overall antitumor efficacy by boosting activation, cytokines secretion, effector T Our findings indicate luteolin, standalone agent, can inhibit proliferation colon lung cancer cells both certain extent. When activated CTLs, it upregulated expression CD25 CD69 resulted higher levels IL-2, TNF-α, IFN-γ secretion vitro. In vivo, this significantly curtailed subcutaneous extended mean time tumor-bearing mice (HCT116, A549), outperforming monotherapy. Furthermore, therapy may be attributable apoptosis cells, reduced decreased YAP expression. The vaccine-activated CTLs presents novel approach for treatment. As adjuvant, downregulates enhancing cytotoxicity, thus improving recognition selective targeting. strategy is promising safe effective
