Water pollution caused by widely used agricultural pesticide chlorpyrifos (CPF) has aroused extensive public concern. While previous studies have reported on toxic effect of CPF aquatic animal, little is known about its common carp (Cyprinus carpio L.) livers. In this experiment, we exposed to (11.6 μg/L) for 15, 30, and 45 days establish a poisoning model. Histological observation, biochemical assay, quantitative real-time polymerase chain reaction (qRT-PCR), Western blot, integrated biomarker response (IBR) were applied assess the hepatotoxicity in carp. Our results displayed that exposure damaged histostructural integrity induced liver injury Furthermore, found CPF-induced may be associated with mitochondrial dysfunction autophagy, as evidenced swollen mitochondria, broken ridges, increased number autophagosomes. Moreover, decreased activities ATPase (Na+/K+-ATPase, Ca2+-ATPase, Mg2+-ATPase, Ca2+Mg2+-ATPase), altered glucose metabolism-related genes (GCK, PCK2, PHKB, GYS2, PGM1, DLAT), activated energy-sensing AMPK, indicating energy metabolism disorder. The activation AMPK further mitophagy via AMPK/Drp1 pathway, autophagy AMPK/mTOR pathway. Additionally, oxidative stress (abnormal levels SOD, GSH, MDA, H2O2) livers, which contributed induction autophagy. Subsequently, confirmed time-dependent IBR assessment. findings presented new insight into molecular mechanism induced-hepatotoxicity carp, provided theoretical basis evaluating toxicity organisms.

Aquatic organisms such as fish can accumulate high concentrations of arsenic (As), which has toxic effects on fish. However, whether the intestinal flora are involved in As damage to tissues and underlying process unclear. Common carp (Cyprinus carpio) were exposed (2.83 mg/L) water for 30 days, blood, muscle, intestine, intestine samples collected. Intestinal pathological sections observed, lipopolysaccharide (LPS) levels serum accumulation tight junction-related factors measured. The gut microbiota was analysed by 16S rRNA sequencing. results showed that treatment decreased abundance microbiota, increased number harmful bacteria, beneficial bacteria intestine. Among resulted a significant (P < 0.05) increase (such Fusobacteriota, Bacteroidota (LPS-producing bacteria), Verrucomicrobiota, Bacteroides, Aeromonas, Stenotrophomonas) decrease Actinobacteriota, Planctomycetota, Firmicutes, Reyranella, Akkermansia, Pseudorhodobacter), further demonstrated affects flora. addition, exposure LPS level bacteria) exhibits six at phylum level, indicates produced serum. Lead (Pb), a hazardous heavy metal, can damage the health of organisms. However, it is not clear whether Pb chicken cerebellums and thalami. Selenium (Se), an essential nutrient for organisms, has palliative effect on poisoning in chickens. In our experiment, model chickens treated with Se alone combination was established to investigate molecular mechanism alleviating Pb-caused both Our morphological results indicated that caused apoptotic lesions, such as mitochondrial nuclear damage. Further, anti-apoptotic gene Bcl-2 decreased; contrary, four pro-apoptotic genes (p53, Bax, Cyt c, Caspase-3) increased under treatment, meaning apoptosis via p53-Cyt c-Caspase-3 pathway.
