Polydatin (PD), a natural product derived from Polygonum cuspidatum, has anti-inflammatory and antioxidant effects significant benefits in treating allergic diseases. However, its role mechanism rhinitis (AR) have not been fully elucidated. Herein, we investigated the effect of PD AR. AR model was established mice with OVA. Human nasal epithelial cells (HNEpCs) were stimulated IL-13. HNEpCs also treated an inhibitor mitochondrial division or transfected siRNA. The levels IgE cellular inflammatory factors examined by enzyme linked immunosorbent assay flow cytometry. expressions PINK1, Parkin, P62, LC3B, NLRP3 inflammasome proteins, apoptosis proteins tissues measured Western blot. We found that suppressed OVA-induced thickening eosinophil accumulation mucosa, reduced IL-4 production NALF, regulated Th1/Th2 balance. In addition, mitophagy induced after OVA challenge IL-13 stimulation. Meanwhile, enhanced PINK1-Parkin-mediated but decreased reactive oxygen species (mtROS) production, activation, apoptosis. PD-induced abrogated PINK1 knockdown Mdivi-1 treatment, indicating key PINK1-Parkin mitophagy. Moreover, damage, mtROS under exposure more severe treatment. Conclusively, may exert protective on promoting mitophagy, which further suppresses tissue damage through decreasing activation.

Introduction Allergic rhinitis (AR) is an upper airway inflammatory disease of the nasal mucosa. Conventional treatments such as symptomatic pharmacotherapy and allergen-specific immunotherapy have considerable limitations drawbacks. As emerging therapy with regenerative potential immunomodulatory effect, mesenchymal stem cell-derived exosomes (MSC-Exos) recently been trialed for treatment various autoimmune diseases. Methods In order to achieve sustained protected release MSC-Exos intranasal administration, we fabricated Poly(lactic-co-glycolic acid) (PLGA) micro nanoparticles-encapsulated (PLGA-Exos) using mechanical double emulsion local AR. Preclinical in vivo imaging, ELISA, qPCR, flow cytometry, immunohistochemical staining, multiomics sequencing were used phenotypic mechanistic evaluation therapeutic effect PLGA-Exos vitro . Results The results showed that our PLGA platform could efficiently encapsulate a manner. At protein level, upregulated IL-2, IL-10 IFN-γ, downregulated IL-4, IL-17 antigen-specific IgE ovalbumin (OVA)-induced AR mice. cellular reduced Th2 cells, increased Tregs, reestablished Th1/Th2 balance. tissue significantly attenuated infiltration immune cells (e.g., eosinophils goblet cells) Finally, analysis discovered several signaling cascades, e.g., peroxisome proliferator-activated receptor (PPAR) pathway glycolysis pathway, might mechanistically support PLGA-Exos. Discussion For first time, present biomaterial-facilitated delivery system novel promising strategy treatment.

Abstract: Patients suffering from allergic rhinitis often attempt to self-manage their symptoms and may seek advice pharmacists about nonprescription product choices. Several drug classes, both prescription over-the-counter (OTC), are available, including intranasal corticosteroids (INCSs); oral, intranasal, ocular antihistamines; leukotriene antagonists; topical systemic decongestants, as well immunotherapies. Selection of the optimal treatment approach depends on temporal pattern, frequency, severity symptoms patient's age. Nasal congestion is typically most bothersome symptom, although rhinorrhea, postnasal drip, also problematic. Together, these adversely impact quality life, work productivity, sleep quality, ability perform daily activities, particularly when uncontrolled. Practice guidelines recognize that INCSs effective medications for controlling symptoms, nasal congestion. Available INCS products have comparable safety efficacy profiles, but they differ in formulation characteristics sensory attributes. barriers can impede use INCSs, concerns safety, misperceptions regarding loss response frequent use, undesirable sensations associated with administration. Given increasing number available OTC, help allay by discussing expectations, recommending favorable characteristics, reviewing proper technique administration selected product. These steps foster a collaborative relationship between patient pharmacist rhinitis. Keywords: allergy, sprays, medications, counseling, pharmacy practice
