Berberine (BBR), an isoquinoline alkaloid extracted from Coptidis Rhizoma, has a long history of treating dysentery in the clinic. Over past two decades, polytrophic, pharmacological, and biochemical properties BBR have been intensively studied. The key functions BBR, including anti-inflammation, antibacterial, antioxidant, anti-obesity, even antitumor, discovered. However, underlying mechanisms BBR-mediated regulation still need to be explored. Given that is also natural nutrition supplement, modulatory effects on nutritional immune responses attracted more attention investigators. In this mini-review, we summarized latest achievements inflammation, gut microbes, macrophage polarization, associated with their possible tools pathogenesis therapy ulcerative colitis cancer recent 5 years. We discuss therapeutic efficacy anti-inflammatory actions benefit future clinical applications.

Drug delivery systems (DDSs) are a multidisciplinary approach toward the effective of drugs to their target sites. Natural small molecule (NSM) compounds with anticancer activity, self-assembly and co-assembly functions show great potential for application as novel DDSs in biomedical field. NSMs widely sourced, have many modification sites, readily form hydrogen bonds, π-π interactions, van der Waals other non-covalent bonds solvents, resulting ordered structures. Moreover, good biocompatibility bioactivity allow compositions based on these be used life science applications such tissue engineering, drug cell imaging, showing medical value DDSs. Abstract Neutrophil extracellular traps (NET), formed by the release of decondensed chromatin and granules, have been shown to promote tumor progression metastasis. Tumor-associated neutrophils in hepatocellular carcinoma (HCC) are prone NET formation, highlighting need for a more comprehensive understanding mechanisms action NETs liver cancer. Here, we showed that DNA (NET-DNA) binds transmembrane coiled-coil domains 6 (TMCO6) on CD8+ T cells impair antitumor immunity thereby HCC progression. TGFβ1 induced which recruited cells. Binding NET-DNA inhibited function while increasing apoptosis secretion, forming positive feedback loop further stimulate formation immunosuppression. Mechanistically, N-terminus TMCO6 interacted with suppressed T-cell receptor signaling NFκB p65 nuclear translocation. Blocking inhibiting PAD4 potent effects wild-type mice but not TMCO6−/− mice. In clinical samples, expressing had an exhausted phenotype. inhibition or deficiency combined anti-PD-1 abolished NET-driven vivo. Lung cancer is the second most prevalent and ranks first in cancer-related death worldwide. Due to resistance development conventional therapy strategies, including chemotherapy, radiotherapy, targeted therapy, immunotherapy, various natural products their extracts have been revealed as alternatives. Berberine (BBR), which present stem, root, bark of trees, could exert anticancer activities by regulating tumor cell proliferation, apoptosis, autophagy, metastasis, angiogenesis, immune responses via modulating several signaling pathways within microenvironment. its poor water solubility, pharmacokinetics/bioavailability profile, extensive p-glycoprotein-dependent efflux, BBR application (pre) clinical studies restricted. To overcome these limitations, can be encapsulated nanoparticle (NP)-based drug delivery systems, monotherapy or combinational improve therapeutic efficacy. Nanoformulations also facilitate selective into lung cells. In addition BBR, especially cancer, here we reviewed nanoformulations, polymeric NPs, metal-based carbon nanostructures, others, treatment cancer.

Neutrophils represent the most abundant cell type of leukocytes in human blood and have been considered a vital player innate immune system first line defense against invading pathogens. Recently, several studies showed that neutrophils play an active role response during cancer development. They exhibited both pro-oncogenic anti-tumor activities under influence various mediators tumor microenvironment. can be divided into subpopulations, thus contradicting traditional concept as homogeneous population with specific function immunity opening new horizons for therapy. Despite promising achievements this field, full understanding tumor–neutrophil interplay is currently lacking. Abstract Lung cancer (LC) is the leading cause of cancer-related deaths globally. It accounts for more than 1.9 million cases each year due to its complex and poorly understood molecular mechanisms that result in unregulated cell proliferation metastasis. β-Catenin a developmentally active protein controls proliferation, metastasis, polarity fate during homeostasis aids progression via epithelial–mesenchymal transition. Therefore, inhibition β-catenin pathway could attenuate LC. Berberine, an isoquinoline alkaloid which known anti-cancer anti-inflammatory properties, demonstrates poor solubility bioavailability. In our study, we have encapsulated berberine into liquid crystalline nanoparticles improve physiochemical functions studied if these target inhibit human lung adenocarcinoma line (A549) at both gene levels. We observed first time 5 µM significantly expression protein. The interaction between was further validated by simulation studies. Targeting with represents promising strategy management progression.
