Inflammatory bowel diseases (IBDs), such as ulcerative colitis (UC) or Crohn's disease, are becoming a growing global problem due to the limitations of current treatments, which fail address needs patients effectively. UC is characterized by widespread inflammation mucosal lining, affecting both rectum and entire length colon. Over past forty years, traditional treatments for IBDs have primarily relied on anti-inflammatory drugs immunosuppressive medications. Treatment could be more effective if specifically targeted act directly Conventional drug delivery systems encounter numerous challenges their way colon, physiological barriers disease severity. To these issues, pH-dependent carriers emerged promising advancement, offering tolerable treatment UC. Epigallocatechin gallate (EGCG) is a polyphenol present in green tea (Camellia sinensis), which has revealed anti-cancer effects toward variety of cancer cells vitro and protective potential against neurodegenerative diseases such as Alzheimer's Parkinson's. Unfortunately, EGCG presents disappointing bioavailability after oral administration, primarily due to its chemical instability poor absorption. Due these limitations, currently not used medication, but only dietary supplement the form extract. Therefore, it needs further modifications before being considered suitable for extensive medical applications. In this article, we review scientific literature about derivatives focusing on their biological properties The most common epigallocatechin rely introducing fatty acid chains or sugar molecules structure modify solubility. Another frequently employed procedure based blocking EGCG's hydroxyl groups with various substituents. Novel reveal interesting properties, which, antioxidant, anti-inflammatory, antitumor antimicrobial, are especially important. It worth noting that promising higher stability activity than base EGCG.

Acute severe ulcerative colitis (ASUC) is a growing health burden that often requires treatment with multiple therapeutic agents. As inflammation localised in the rectum and colon, local drug delivery using suppositories could improve outcomes. Three-dimensional (3D) printing novel manufacturing tool permits combination of drugs personalised dosage forms, created based on each patient's disease condition. This study, for first time, demonstrates feasibility producing 3D printed two anti-inflammatory agents, budesonide tofacitinib citrate, ASUC. both are poorly water-soluble, suppositories' ability to self-emulsify was exploited their performance. The were fabricated via semi-solid extrusion (SSE) contained citrate varying doses (10 or 5 mg; 4 2 mg, respectively). displayed similar dissolution disintegration behaviours irrespective content, demonstrating flexibility technology. Overall, this study SSE create multi-drug ASUC, possibility titrating progression.

Infliximab is a monoclonal antibody that plays an important role in the management and treatment of chronic inflammatory bowel diseases (IBD). Due to its macromolecular structure, delivery through oral route challenging, limiting administration only via parenteral route. The rectal offers alternative way for administering infliximab, allowing it be localised at disease site circumventing passage across alimentary canal thus, maintaining integrity bioactivity. Three-dimensional (3D) printing advanced production technology permits creation dose-flexible drug products from digital designs. current study assessed feasibility utilising semi-solid extrusion 3D fabrication infliximab-loaded suppositories local IBD. Various inks composed Gelucire® (48/16 or 44/14) mixed with coconut oil and/or purified water were investigated. It was shown following reconstitution water, infliximab solution can directly incorporated into ink 48/16 withstand process, resulting well-defined suppositories. Since content temperature are critical safeguarding infliximab's potency, effect changing composition parameters on biologic efficiency evaluated by measuring binding capacity (i.e., amount actively binds antigen exert effect). Despite loading assays showing remains intact printing, found incorporation isolation results ∼65% capacity. However, when added mixture, increases up ∼85%. These promising demonstrate has potential exploited as novel platform fabricating dosage forms containing biopharmaceuticals, avoiding patients' compliance issues observed injectables addressing their unmet needs.
