Intestinal mucosal injury may arise from various factors. While many drugs target the causative factors, none directly stimulate wound healing. We found that specific focal adhesion kinase (FAK) activator, M64HCl, promotes intestinal healing in mice. This study aims to further validate therapeutic impact of M64HCl on repair rats as a second species. Wistar were assigned one four groups: normal control, 1-day + vehicle, 4-day or M64HCl. was induced by serosally applying 75% acetic acid. Immediately after injury, received either continuous infusion (25 mg/kg/day) its vehicle (saline). Four days post-injury, blood drawn measure levels and assess liver kidney function. The intestines removed opened, ulcer areas photographed for size quantification, tissues fixed histological immunohistochemical analysis. substantially reduced area gross examination, while analysis showed alleviation pathological changes with treatment. ABSTRACT Porcine epidemic diarrhea virus (PEDV) causes severe intestinal damage and high mortality in neonatal piglets. The continuous emergence of new strains has brought challenges to prevention control. In this study, we isolated characterized a prevalent PEDV virulent strain analyzed 19,612 jejunal cells from PEDV-infected control piglets using single-cell sequencing, revealing significant changes cellular composition, gene expression, intercellular communication. response infection, epithelial repair was enhanced through increased proliferation differentiation stem cells, transit-amplifying (TA) progenitor into enterocytes. Additionally, disrupted communication, compromising functionality while triggering immune responses, with IFN-γ IL-10 signaling activation acting as critical regulators balance tissue homeostasis.
