Abstract Alzheimer's disease is characterized by memory loss and pathological changes in the brain such as amyloid beta tau pathology, well irregular neural network activity. We studied how intranasal insulin impacts oscillations an rat model induced STZ. Male Wistar rats were subjected to intracerebroventricular STZ injection, followed therapy. Electrophysiological recordings conducted hippocampus medial prefrontal cortex assess local field potentials, elucidating insignificant alterations theta-gamma following treatment. However, administration ameliorated deficits hippocampal sharp-wave ripples augmented hippocampal-prefrontal theta coherence. Concurrently, therapy conferred enhancements spatial object recognition performance behavioral tests. Furthermore, mitigated protein pathology neuronal loss. Our findings underscore potential of enhance function via modulating cortical synchronization alleviating impairments ripples.

Chronic cerebral hypoperfusion (CCH) is increasingly recognized as a common cognitive impairment-causing mechanism. However, no clinically effective drugs to treat impairment due CCH have been identified. An abnormal distribution of neural oscillations was found in the hippocampus rats. By releasing various neurotransmitters, distinct afferent fibers influence neuronal hippocampus. Enriched environments (EE) are known improve levels by modulating neurotransmitter homeostasis. Using EE an intervention, we examined three classical neurotransmitters and dynamics rat model. The results showed that significantly improved balance (acetylcholine, glutamate, GABA) hippocampus, enhanced strength theta slow-gamma (SG) rhythms, dramatically coupling across frequency bands Furthermore, expression vesicular transporters-vesicular acetylcholine transporters (VAChT) GABA (VGAT)-was reduced rats, whereas glutamate transporter 1 (VGLUT1) abnormally elevated. partially restored protein maintain hippocampal neurotransmitters. More importantly, causal mediation analysis increased power rhythm increasing level VAChT VGAT, which then phase amplitude theta-SG finally led improvement CCH. These findings shed light on role disruption oscillations. This study has implications for our knowledge disease pathways.
