Survival depends on a balance between seeking rewards and avoiding potential threats, but the neural circuits that regulate this motivational conflict remain largely unknown. Using an approach-food vs. avoid-predator threat test in rats, we identified subpopulation of neurons anterior portion paraventricular thalamic nucleus (aPVT) which express corticotrophin-releasing factor (CRF) are preferentially recruited during conflict. To maintain energy homeostasis, orexigenic (appetite-inducing) and anorexigenic (appetite suppressing) brain systems functionally interact to regulate food intake. Within the hypothalamus, neurons that express agouti-related protein (AgRP) sense factors orchestrate an increase in food-seeking behavior. In contrast, calcitonin gene-related peptide (CGRP)-expressing parabrachial nucleus (PBN) suppress feeding. PBN CGRP become active response hormones released following a meal, including amylin, secreted by pancreas, cholecystokinin (CCK), small intestine.
