Abstract Understanding the long-term effects of stress on brain function is crucial for understanding mechanisms depression. The BALB/c mouse strain has high susceptibility to and thus an effective model repeated social defeat (SDS) mice, however, are not clear. Here, we investigated SDS in male mice over subsequent two weeks. Some defeated immediately exhibited avoidance, whereas anxiety-like behavior was only evident at later periods. Furthermore, segregated into groups based level namely, avoidant nonavoidant mice. characteristic avoidance or nonavoidance each individual fixed In addition, developed a semi-automated method analyzing c-Fos expression investigate effect activity more than weeks after end exposure. Following interaction, reduced several regions compared with control correlation among these areas, exception medial prefrontal cortex (mPFC) central amygdala (CeA), increased suggesting synchrony. Abstract Neuropsychiatric disorders, such as major depression, anxiety and post-traumatic stress disorder, tend to be long-term conditions in whose development maintenance are central pathogenic factors. Translational mouse models widely used neuropsychiatric research, exploiting social non-social stressors investigate the mechanisms underlying their detrimental effects. However, most studies focus on short-term consequences of chronic stress, whereas only a few interested course. This is counterintuitive given human that preclinical designed mimic. In this review, we have summarized limited work date effects mice models. First, different presented definition short- vs. sequelae proposed. On basis, behavioral, endocrine, vegetative addressed before examining data cellular molecular alterations brain. Finally, future directions for research discussed.

Abstract Optogenetic methods provide efficient cell-specific modulations, and the ability of simultaneous neural activation inhibition in same brain region freely moving animals is highly desirable. Here we report bidirectional neuronal activity manipulation accomplished by a wireless, dual-color optogenetic probe synergy with co-expression two spectrally distinct opsins (ChrimsonR stGtACR2) rodent model. The flexible comprises vertically assembled, thin-film microscale light-emitting diodes lateral dimension 125 × 180 µm 2 , showing colocalized red blue emissions enabling chronic vivo operations desirable biocompatibilities. Red or irradiations deterministically evoke silence neurons co-expressing opsins. interferes dopaminergic ventral tegmental area mice, increasing decreasing dopamine levels. Such regulations further generate rewarding aversive behaviors interrogate social interactions among multiple mice. These technologies create numerous opportunities implications for research.

The ability to recognize motivationally salient events and adaptively respond them is critical for survival. Here, we tested whether dopamine (DA) neurons in the dorsal raphe nucleus (DRN) contribute this process both male female mice. Population recordings of DRN DA during associative learning tasks showed that their activity dynamically tracks motivational salience, developing excitation reward-paired shock-paired cues. response reward-predicting cues was diminished after satiety, suggesting modulation by internal states. also greater unexpected outcomes than expected outcomes. Two-photon imaging demonstrated majority individual developed activation reward but not shock-predicting cues, which surprising qualitatively distinct from population results. Performing same fear procedures freely-moving head-fixed groups revealed head-fixation itself abolished neural aversive indicating its behavioral context. Overall, these results suggest encode dependent on external factors.
