Episodic memories are encoded by experience-activated neuronal ensembles that remain necessary and sufficient for recall. However, the temporal evolution of memory engrams after initial encoding is unclear. In this study, we employed computational experimental approaches to examine how neural composition selectivity change with consolidation. Our spiking network model yielded testable predictions: transition from unselective selective as neurons drop out into engrams; inhibitory activity during recall essential selectivity; synaptic plasticity consolidation critical become selective. Using activity-dependent labeling, longitudinal calcium imaging a combination optogenetic chemogenetic manipulations in mouse dentate gyrus, conducted contextual fear conditioning experiments supported our model's predictions. results reveal dynamic changes engram mediated crucial emergence selectivity.

Abstract Complex experimental protocols often require multi-modal data acquisition with precisely aligned timing, as well state- and behavior-dependent interventions. Tailored solutions are mostly restricted to individual setups lack flexibility interoperability. We present an open-source, Linux-based integrated software solution, called ‘Syntalos’, for simultaneous synchronization of from arbitrary number sources, including multi-channel electrophysiological recordings different live imaging devices, closed-loop, real-time interventions actuators. Precisely matching timestamps all inputs ensured by continuous statistical analysis correction devices’ timestamps. New sources can be minimal programming skills. Data is stored in a comprehensively structured format facilitate pooling or sharing between laboratories. Syntalos enables synchronized closed-loop multiple approaches. Preliminary neuroscientific experiments on mice research questions show the successful performance easy-to-learn structure suite.

Abstract Behavior can be remarkably consistent, even over extended time periods, yet whether this is reflected in stable or ‘drifting’ neuronal responses to task features remains controversial. Here, we find a persistently active ensemble of neurons the medial prefrontal cortex (mPFC) mice that reliably maintains trajectory-specific tuning several weeks while performing an olfaction-guided spatial memory task. This task-specific reference frame stabilized during learning, upon which repeatedly show little representational drift and maintain their across long pauses exposure repeated changes cue-target location pairings. These data thus suggest ‘core ensemble’ forming task-relevant space for performance consistent behavior periods time.

Representational drift refers to the dynamic nature of neural representations in brain despite behavior being seemingly stable.
