Background The retrograde endocannabinoid (eCB) pathway is closely associated with the etiology of major depressive disorder (MDD) at both pathophysiological and genetic levels. This study aimed to investigate potential role mutations in eCB underlying mechanisms Han Chinese patients MDD. Methods A total 96 drug-naïve first-episode MDD 62 healthy controls (HCs) were recruited. Whole-exome sequencing was performed identify gene mutation profiles Results filtered focus on low-frequency variants rare (minor allele frequencies <0.05) related phenotypes. Enrichment analyses for 146 selected genes examine pathways which most significant enrichment occurred. protein–protein interaction (PPI) network analysis explore biological functions pathway. Finally, based current literature, a preliminary conducted effect function this Our identified (15.02%) depression-related when compared HCs, 37 enriched signaling Seven hub mitochondrial function, including Complex I (NDUFS4, NDUFV2, NDUFA2, NDUFA12, NDUFB11) protein (PARK7) enzyme (DLD) regulation oxidative stress. Conclusion These results indicate that represent etiological factors pathogenesis

Some of the most prevalent neurodegenerative disorders, including Alzheimer’s and Parkinson’s disease, are proteinopathies characterized by accumulation specific protein aggregates in brain. Such misfolded can trigger modulation innate adaptive immune systems subsequently lead to chronic neuroinflammation that drives onset progression diseases. Since there is still no effective disease-modifying treatment, new therapeutic targets for have been sought. The endocannabinoid system, particular cannabinoid CB2 receptors, extensively studied, due their important role neuroinflammation, especially microglial cells. Several studies shown promising effects receptor activation on reducing aggregation-based pathology as well attenuating inflammation several dementia-related symptoms. In this review, we discuss available data receptors potential benefits limitations agonists these therapy proteinopathies.

Behaviour is the response of living things to their environment and external stimulation, one parameters be observed when assessing animal welfare. Any alteration from conditions found in nature can lead occurrence some specific behaviours, called stereotypies which are characterised as repetitive, consistent patterns behaviour usually defined having no apparent ultimate or proximal functions. Abstract Introduction Repetitive transcranial magnetic stimulation (rTMS) is a clinically useful therapy for depression. However, the effects of rTMS on metabolism fatty acids (FAs) and composition gut microbiota in depression are not well established. Methods Mice received (15 Hz, 1.26 T) seven consecutive days after exposure to chronic unpredictable mild stress (CUMS). The subsequent depressive‐like behaviors, stool samples, as medium‐ long‐chain (MLCFAs) plasma, prefrontal cortex (PFC), hippocampus (HPC) were evaluated. Results CUMS induced remarkable changes microbiotas acids, specifically community diversity PUFAs brain. 15 Hz treatment alleviates behaviors partially normalized alterations MLCFAs, especially abundance Cyanobacteria , Actinobacteriota levels polyunsaturated (PUFAs) PFC. Conclusion These findings revealed that modulation might partly contribute antidepressant effect rTMS.

Abstract Periprosthetic osteolysis (PPO) induced by wear particles at the interface between prosthesis and bone is a crucial issue of periprosthetic loss implant failure. After tear, granular material accumulates around joint prosthesis, causing chronic inflammatory response, progressive osteoclast activation eventual loosening prosthesis. Although many studies have been conducted to address after replacement surgeries, they not fully addressed these issues. Focusing on has important theoretical implications. Cannabinoid type II receptor (CB2) seven-transmembrane that predominantly distributed in human immune system revealed be highly expressed bone-associated cells. Previous shown modulation CB2 positive effect metabolism. However, exact mechanism yet elucidated. In our experiments, we found NOX1-mediated ROS accumulation was involved titanium particle-stimulated differentiation. Furthermore, confirmed blockade alleviated inhibiting oxidative stress pathway. animal downregulation occurrence particle-induced cranial osteoclasts scavenging intracellular ROS. Collectively, results suggest may an attractive promising therapeutic scheme for differentiation preventing PPO.

Despite decades of rigorous research and numerous clinical trials, Alzheimer’s disease (AD) stands as a notable healthcare challenge this century, with effective therapeutic solutions remaining elusive. Recently, the endocannabinoid system (ECS) has emerged an essential target due to its regulatory role in different physiological processes, such neuroprotection, modulation inflammation, synaptic plasticity. This aligns previous showing that cannabinoid receptor ligands have potential trigger functional structure neuronal brain networks, potentially impacting memory processing. Therefore, our study aims assess effects prolonged, intermittent exposure (over 90 days) JWH-133 (0.2 mg/kg) EU-GMP certified Cannabis sativa L.
