The intricate mechanisms governing homeostasis are substantially impacted extracellular vesicles (EVs), play crucial roles in both pathological physiological contexts. EVs derived from various sources exert distinct effects on osteoporosis. Specifically, released osteoblasts, endothelial cells, myocytes, mesenchymal stem cells contribute formation due their unique cargo proteins, miRNAs, cytokines. Conversely, secreted osteoclasts immune promote resorption inhibit formation. Furthermore, use as therapeutic modalities or biomaterials for diagnosing managing osteoporosis promising. Here, we review current understanding impact homeostasis, including classification biogenesis regulatory present an overview latest research progress treating using EVs. Glioblastoma (GB) is a rare but extremely aggressive brain tumor that significantly impacts patient outcomes, affecting both duration and quality of life. The protocol established by Stupp colleagues in 2005, based on radiotherapy chemotherapy with Temozolomide, following maximum safe surgical resection remains the gold standard for GB treatment; however, it evident nowadays extreme intratumoral intertumoral heterogeneity, as well invasiveness tendency to recur, are not compatible routine unfortunately ineffective treatment. This review article summarizes main challenges search new valuable therapies focuses impact extracellular vesicle (EV) research exploitation may have field. EVs natural particles delimited lipidic bilayer filled functional cellular content released uptaken cells key means cell communication. Furthermore, stable body fluids tolerated immune system, able cross physiological, interspecies, interkingdom barriers target specific cells, releasing inherent or externally loaded functionally active molecules. Abstract Cancer, a prevalent and complex disease, presents significant challenge to the medical community. It is characterized by irregular cell differentiation, excessive proliferation, uncontrolled growth, invasion of nearby tissues, spread distant organs. Its progression involves interplay several elements processes. Extracellular vesicles (EVs) serve as critical intermediaries in intercellular communication, transporting molecules such lipids, RNA, membrane, cytoplasmic proteins between cells. They significantly contribute progression, development, dissemination primary tumors facilitating exchange information transmitting signals that regulate tumor growth metastasis. However, EVs do not have singular impact on cancer; instead, they play multifaceted dual role. Under specific circumstances, can impede influence cancer delivering oncogenic factors or triggering an immune response. Furthermore, from different sources demonstrate distinct advantages inhibiting cancer. This research examines biological characteristics their involvement development establish theoretical foundation for better understanding connection Here, we discuss potential various therapy, well current status future prospects engineered developing more effective treatments.

Neutrophils represent the most abundant cell type of leukocytes in human blood and have been considered a vital player innate immune system first line defense against invading pathogens. Recently, several studies showed that neutrophils play an active role response during cancer development. They exhibited both pro-oncogenic anti-tumor activities under influence various mediators tumor microenvironment. can be divided into subpopulations, thus contradicting traditional concept as homogeneous population with specific function immunity opening new horizons for therapy. Despite promising achievements this field, full understanding tumor–neutrophil interplay is currently lacking. In review, we try to summarize current view on neutrophil heterogeneity cancer, discuss different communication pathways between tumors neutrophils, focus implementation these findings develop neutrophil-based therapies.

Tumor cells actively incorporate molecules (e.g., proteins, lipids, RNA) into particles named extracellular vesicles (EVs). Several groups have demonstrated that EVs can be transferred to target (recipient) cells, making an important means of intercellular communication. Indeed, are able modulate the functions by reprogramming signaling pathways. In a cancer context, promote formation supportive tumor microenvironment (TME) and (pre)metastatic niches. Recent studies revealed immune their secretome, including EVs, changes in TME immunosuppressive natural killer, dendritic T B monocytes, macrophages) allow establish propagate. Despite growing knowledge on roles as modulators response/escape, translation clinical practice remains its early stages, hence requiring improved translational research field. Here, we comprehensively review current most recent evasion immunosuppression both solid tumors hematological malignancies. We also highlight utility EV-mediated targeting EV-engineering. Abstract Background Tumor-derived small extracellular vesicles (sEVs) can promote tumorigenic and metastatic capacities in less aggressive recipient cells mainly through the biomolecules their cargo. However, despite recent advances, specific molecules orchestrating these changes are not completely defined. Lactadherin is a secreted glycoprotein typically found milk fat globule membrane. Its overexpression has been associated with increased tumorigenesis metastasis breast cancer (BC) other tumors. neither its presence sEVs by BC cells, nor role sEV-mediated intercellular communication have described. The present study focused on of lactadherin-containing from MDA-MB-231 triple-negative (TNBC) (sEV-MDA231) promotion pro-metastatic non-tumorigenic non-metastatic vitro, as well murine model peritoneal carcinomatosis. Results We show that lactadherin it higher sEV-MDA231 compared cell-secreted measured ELISA. Incubation increases migration and, to some extent, tumoroid formation capacity but anchorage-independent growth. Remarkably, blockade results significant decrease those vitro. Similarly, intraperitoneally treatment mice greatly increase malignant ascites tumor micronodules, effects were significantly inhibited when was previously blocked sEV-MDA231. Conclusions As our knowledge, provides first evidence promoter of: (i) ii) nodules. These understanding which be used therapeutic option for malignancies.

On other hand, mesenchymal stem cell (MSC) therapy, which is being employed regenerative medicine owing to ability MSCs differentiate into various cells, due humoral factors, including messenger RNA (mRNA), miRNAs, proteins, lipids, encapsulated exosomes derived transplanted cells. New treatments that advocate cell-free therapy using MSC-derived will significantly improve clinical practice. Therefore, highly purified perform their original functions desirable.
