Ageing is becoming an increasingly serious problem; therefore, there urgent need to find safe and effective anti-ageing drugs.AimsTo investigate the effects of Bazi Bushen capsule (BZBS) on senescence mesenchymal stem cells (MSCs) explore its mechanism action.MethodsNetwork pharmacology was used predict targets BZBS in delaying MSCs. For vitro studies, MSCs were treated with D-gal, BZBS, NMN, cell viability, senescence, stemness-related genes, cycle studied using counting kit-8 (CCK-8) assay, SA-β-galactosidase (SA-β-gal) staining, Quantitative Real-Time PCR (qPCR) flow cytometry (FCM), respectively. The objective of this study is to comprehensively identify the core pharmacological components and their respective targets three medicinal fungi Sanghuangs including Sanghuangporus vaninii (SV), lonicericola (SL), Inonotus hispidus (IH). ABSTRACT Gancao Fuzi decoction (GCFZT) is a traditional Chinese formula, which has been commonly used in clinical practice to treat inflammatory diseases. However, the active substance of GCFZT treatment inflammation not fully clarified. In this study, we orthogonal experiments design different formulations, resulting total 16 formulations. Subsequently, UPLC‐Q‐TOF‐MS/MS was analyze chemical composition and anti‐inflammatory activity differences these formulations were evaluated through an LPS‐induced RAW264.7 cell model. Combined with machine learning algorithms such as PLS‐DA RF, four main substances screened. Finally, network pharmacology techniques investigate potential mechanisms substances, results showed that mainly regulates expression core targets ALOX5, NFKB1, TLR4 chlorogenic acid, riboflavin, formononetin, thereby affecting NF kappa B signaling pathway, Toll‐like receptor Th17 differentiation. This study provides reference for mechanism scientific basis its application.
