The therapeutic effect of traditional chemodynamic therapy (CDT) agents is severely restricted by their weakly acidic pH and glutathione (GSH) overexpression in the tumor microenvironment. Here, fusiform-like copper(II)-based tetrakis(4-carboxy phenyl)porphyrin (TCPP) nanoscale metal–organic frameworks (nMOFs) were designed constructed for first time (named PCN-224(Cu)-GOD@MnO2). coated MnO2 layer can not only avoid conjugation glucose oxidase (GOD) to damage normal cells but also catalyzes generation O2 from H2O2 enhance oxidation (Glu) GOD, which provides abundant subsequent Cu+-based Fenton-like reaction. Meanwhile, Cu2+ chelated TCPP ligand converted Cu+ excess GSH tumor, reduces antioxidant activity improve CDT effect. Next, reacts with plentiful enzyme catalysis produce a toxic hydroxyl radical (•OH), singlet oxygen (1O2) synchronously generated combination Cu+, O2, H2O via Russell mechanism. Furthermore, nanoplatform be used both TCPP-based vivo fluorescence imaging Mn2+-induced T1-weighted magnetic resonance imaging. In conclusion, PCN-224(Cu)-GOD@MnO2 nMOFs facilitate efficiency starvation relief hypoxia depletion after acting as an accurate guide.

Light-sensitive yolk–shell nanoparticles (YSNs) as remote-controlled and stimuli-responsive theranostic platforms provide an attractive method for synergistic cancer therapy. Herein, a kind of novel multifunctional YSNs has been successfully constructed by integrating star-shaped gold (Au star) the second near-infrared (NIR-II) photothermal yolks biodegradable crystalline zeolitic imidazolate framework-8 (ZIF-8) shells. Development of efficient therapeutic strategy to incorporate ultrasound (US)-triggered sonodynamic therapy (SDT) and ferroptosis is highly promising in cancer therapy. However, the SDT efficacy severely limited by hypoxia high glutathione (GSH) tumor microenvironment, associated with reactive oxygen species (ROS) GSH depletion. Methods: A manganese porphyrin-based metal-organic framework (Mn-MOF) was constructed as a nanosensitizer self-supply (O2) decrease for enhanced ferroptosis. In vitro vivo analysis, including characterization, O2 generation, depletion, ROS lipid peroxidation, antitumor immune microenvironment were systematically evaluated. Results: Mn-MOF exhibited catalase-like decreasing activity vitro. After internalization into cells, persistently catalyzed tumor-overexpressed H2O2 in-situ produce relieve GPX4, which facilitated formation kill cells upon US irradiation hypoxic tumors. Abstract Despite the promise of sonodynamic processes in cancer therapy, existing sonosensitizers often fail to regulate generation reactive oxygen species (ROS) against tumors, potentially leading off‐target toxicity normal tissues. We report a transformable core‐shell nanosonosensitizer (TiO 2 @CaP) that reinvigorates ROS and dissolves its CaP shell release Ca 2+ an acidic tumor microenvironment (TME) under ultrasound activation. Thus, TiO @CaP acts as smart specifically induces mitochondrial dysfunction via overloading intracellular ions synergize with process TME. substantially enhances immunogenic cell death, resulting enhanced T‐cell recruitment infiltration into cold (4T1). In conjunction checkpoint blockade therapy (anti‐PD 1), @CaP‐mediated elicits systemic antitumor immunity, regression non‐treated distant tumors inhibition lung metastasis. This work paves way development “smart” TME‐activatable temporospatial control over responses.

Janus nanoparticles (JNPs) refer to the integration of two or more chemically discrepant composites into one structure system. Studies JNPs have been significant interest due their interesting characteristics stemming from asymmetric structures, which can integrate different functional properties and perform synergetic functions simultaneously. Herein, we present recent progress particles, comprehensively detailing fabrication strategies applications. First, classification is divided three blocks, consisting polymeric composites, inorganic hybrid polymeric/inorganic composites. Then, are alternately summarized, examining self-assembly strategy, phase separation seed-mediated polymerization, microfluidic preparation nucleation growth methods, masking methods. Finally, various intriguing applications presented, including solid surfactants agents, micro/nanomotors, biomedical such as biosensing, controlled drug delivery, bioimaging, cancer therapy, combined theranostics. Furthermore, challenges future works in this field provided.
