Extracellular vesicles (EVs) are present in blood at much lower concentrations (5-6 orders of magnitude) compared to lipoprotein particles (LP). Because LP and EV overlap size density, isolating high-purity EVs is a significant challenge. While the current two-step sequential isolation process using size-expression chromatography (SEC) followed by density gradient (DG) achieves high purity, time-consuming ultracentrifugation (UC) step DG hinders workflow efficiency. This paper introduces an optimized magnetic bead reagent, LipoMin, functionalized with glycosaminoglycans (GAGs), as rapid alternative for removal during second-step about 10 minutes. We evaluated LipoMin's efficacy on two sample types: (a) fractions isolated exclusion (SEC + LipoMin) (b) pellet obtained from (UC LipoMin). The remarkably simple, involving min incubation LipoMin separation LP-depleted EV-containing supernatant. Results enzyme-linked immunosorbent assay (ELISA) revealed that removes 98.2% ApoB SEC fractions, comparable ability process. Importantly, yield (CD81 ELISA) remained 93.0% Western blot analysis confirmed key markers, flotillin CD81, were not compromised. Recombinant (rEV), reference standard, was spiked into recovered 89% CD81 protein. For UC ApoA1 decreased 76.5% while retaining 90.7% CD81. Notably, both colorectal cancer (CRC) Alzheimer's disease (AD) samples processed displayed clear expression relevant clinical markers. With (resulting 96% time saving traditional method), reagent offers efficient depletion, paving way streamlined

Abstract Polymerase chain reaction (PCR)‐based nucleic acid testing has played a critical role in disease diagnostics, pathogen surveillance, and many more. However, this method requires long turnaround time, expensive equipment, trained personnel, limiting its widespread availability diagnostic capacity. On the other hand, clustered regularly interspaced short palindromic repeats (CRISPR) technology recently demonstrated capability for detection with high sensitivity specificity. CRISPR‐mediated biosensing holds great promise revolutionizing procedures developing point‐of‐care diagnostics. This review focuses on recent developments both fundamental CRISPR biochemistry CRISPR‐based techniques. Four ongoing research hotspots molecular diagnostics‐target preamplification‐free detection, microRNA (miRNA) testing, non‐nucleic‐acid SARS‐CoV‐2 detection‐are also covered.

Cancer is a complex deadly disease that has caused global health crisis in recent epochs. Colorectal cancer (CRC) the third most common malignant gastrointestinal disease. It led to high mortality due early diagnostic failure. Extracellular vesicles (EVs) come with promising solutions for CRC. Exosomes (a subpopulation of EVs) play vital role as signaling molecules CRC tumor microenvironment. secreted from all active cells. Exosome-based molecular transport (DNA, RNA, proteins, lipids, etc.) transforms recipient cell's nature. In CRC, cell-derived exosomes (TEXs) regulate multiple events development and progression such immunogenic suppression, angiogenesis, epithelial–mesenchymal transitions (EMT), physical changes extracellular matrix (ECM), metastasis. Biofluid-circulated tumor-derived are potential tool liquid biopsy. colorectal detection creates great impact biomarker research. The exosome-associated theranostics approach state-of-the-art method. this review, we address associated progression, on screening (diagnostic prognostic biomarkers), also highlight several clinical trials, well future directions exosome-based Hopefully, it will encourage researchers develop theranostic fight

Cancer is an unsolved health crisis worldwide. Extracellular vesicles (EVs) address this problem in a new way. In cancer, early detection highly challenging, exosomes (a subpopulation of EVs, originating from endosomes) overcome limitation. tumor-derived (TEXs) play role as signaling molecules cancer development and progression. TEXs required more detailed investigation for specific biomarkers research. Exosome heterogeneity (some the facts regulate it such exosome size, origin, inner molecular diversity) made complication liquid biopsy.
