Abstract Regio‐isomers are utilized to design innovative AIE luminogens (AIEgens) by regulating molecular aggregation behavior. However, relevant examples limited, and the underlying mechanism is not fully understood. Herein, a regio‐isomer strategy used develop AIEgens precisely intermolecular interactions in solid state. Among regio‐isomers it investigated, ortho‐ isomer ( DCM‐O3‐O7 ) exhibits enhanced AIE‐activity than para ‐ DCM‐P6 ), size of ortho substituents crucial for performance. The revealed using DFT calculations single‐crystal analysis. Dual hydrogen bonds (C─H∙∙∙π C─H∙∙∙N) generated between molecules, which contributes form dimers, tetramers, 1D supramolecular structures crystal. By restricting intramolecular motion attenuating π – interactions, solid‐state fluorescence significantly enhanced. This strategy's effectiveness validated other donor–acceptor fluorophores, with DCM‐O6 its analogues serving as efficient probes bioimaging applications. Notably, DCM‐OM , bears morpholinyl instead piperidinyl group, displayed strong lysosome‐targeting ability photostability; DCM‐OP incorporated hydrophilic quaternary ammonium exhibited wash‐free imaging cell membrane‐targeting capabilities; nanoparticles enabled high‐fidelity vivo tumor imaging. Abstract Manufacturing whole cancer cell vaccines (WCCV) with both biosafety and efficacy is crucial for tumor immunotherapy. Pyroptotic cells, due to their highly immunogenic properties, present a promising avenue the development of WCCV. However, successful WCCV based on pyroptotic cells yet be accomplished. Here, facile strategy that utilized photocatalytic carbon dots (CDs) induce pyroptosis fabricating reported. Photocatalytic CDs are capable generating substantial amounts hydroxyl radicals can effectively decrease cytoplasmic pH values under white light irradiation. This process efficiently triggers through reactive oxygen species (ROS)‐mitochondria‐caspase 3‐gasdermin E pathway proton motive force‐driven mitochondrial ATP synthesis pathway. Moreover, in vitro, these CDs‐induced (PCIP) hyperactivate macrophage (M0–M1) upregulation major histocompatibility complex class II expression. In vivo, PCIP induced specific immune‐preventive effects melanoma breast mouse models anticancer immune memory, demonstrating effective work provides novel insights inducing bridges gap fabrication cells.

Abstract Cancer immunotherapy has the potential to revolutionize treatment of malignant tumors, but its effectiveness is limited by low immune response rate and immune‐related adverse events. Pyroptosis, as an inflammatory programmed cell death type, triggers strong acute antitumor immunity, converting “cold” tumors “hot”. Particularly, biomaterials loading pyroptosis inducers targeting tumor microenvironment engineer pyroptosis, have achieved great progress in recent years. Herein, design strategy, mechanism pathway, role induce cancer are comprehensively reviewed. The present review focuses on application biomaterials‐induced immunotherapy, including nanogel, polymer prodrug, nanovesicle, mesoporous material. Network interpenetration plays a crucial role in functionalizing porous framework materials. However, controlling the degree of covalent organic frameworks (COFs) to influence their pore sizes, shapes, and functionalities still remains significant challenge. Here, we demonstrate steric tuning strategy control COF modulate physicochemical properties. By imine condensations 1,1′-bi-2-naphthol-derived tetraaldehydes bearing different alkyl substituents with monomer tetra(p-aminophenyl)-methane, synthesized characterized family two-component three-component chiral COFs interpenetrated dia networks. The groups are periodically appended on walls, types/contents that can be synthetically tuned by minimizing repulsive interactions between groups. Specifically, –OH adopts an dia-c5 topology, those –OMe/–OEt take dia-c4 whereas bulky –OnPr/–OnBu exhibit noninterpenetrated dia-c1 topology. multivariate both –OnBu display either or depending proportion extent significantly affects porosity, thermal stability, chemical resulting varying selective performances adsorption separation dyes asymmetric catalysis.
