Perturbations lipid are responsible for the development various pathologies comprising metabolic syndrome, obesity, type 2 diabetes. Accumulating evidence suggests that circadian oscillators, operative most cells our body, coordinate temporal homeostasis. In this review we summarize current knowledge on regulation digestion, absorption, transportation, biosynthesis, catabolism, storage. Specifically, focus molecular interactions between functional clockwork biosynthetic pathways major classes cholesterol, fatty acids, triacylglycerols, glycerophospholipids, glycosphingolipids, sphingomyelins. A growing body epidemiological studies associate a socially imposed misalignment common modern society incidence disorders, however disruption rhythms connection has only been recently revealed. Here, highlight recent unravel mechanistic link intracellular clocks, homeostasis diseases based animal models clock innovative translational humans. Abstract Ischaemic heart disease represents the leading cause of morbidity and mortality, typically induced by detrimental effects risk factors on cardiovascular system. Although preventive interventions tackling conventional have helped to reduce incidence ischaemic disease, it remains a major death worldwide. Thus, attention is now shifting non-traditional in built, natural, social environments that collectively contribute substantially burden perpetuate residual risk. Of importance, these complex interact non-linearly unpredictable ways often enhance attributable single or collection factors. For this reason, new paradigm called ‘exposome’ has recently been introduced epidemiologists order define totality exposure The purpose review outline how emerging may occurrence with particular impact long-term different environmental pollutants, socioeconomic psychological factors, along infectious diseases such as influenza COVID-19. Moreover, potential mitigation strategies for both individuals communities will be discussed.

Circadian clocks are biological timing mechanisms that generate 24-h rhythms of physiology and behavior, exemplified by cycles sleep/wake, hormone release, metabolism. The adaptive value is evident when internal body daily environmental mismatched, such as in the case shift work jet lag or even mistimed eating, all which associated with physiological disruption disease. Studies animal human models have also unraveled an important role functional circadian modulating cellular organismal responses to cues (ex., food intake, exercise), pathological insults (e.g. virus parasite infections), medical interventions medication). Abstract Aims We estimated the association between outdoor light at night residence and risk of coronary heart disease (CHD) within a prospective cohort older adults in Hong Kong. Methods results Over median 11 years follow-up, we identified 3772 incident CHD hospitalizations 1695 deaths. Annual levels participants’ residential addresses were using time-varying satellite data for composite persistent night-time illumination ∼1 km2 scale. used Cox proportional hazards models to estimate hazard ratios (HRs) 95% confidence intervals (CIs) CHD. The hospitalization mortality exhibited monotonic exposure-response function. An interquartile range (IQR) (60.0 nW/cm2/sr) increase was associated with an HR 1.11 (95% CI: 1.03, 1.18) 1.10 1.00, 1.22) deaths after adjusting both individual area-level factors. did not vary across strata hypothesized Conclusion Among adults, higher caution against causal interpretation these novel findings. Future studies more detailed information on exposure, adaptive behaviours, potential mediators are warranted further examine relationship risk.

Every organism has an intrinsic biological rhythm that orchestrates processes in adjusting to daily environmental changes. Circadian rhythms are maintained by networks of molecular clocks throughout the core and peripheral tissues, including immune cells, blood vessels, perivascular adipose tissues. Recent findings have suggested strong correlations between circadian clock cardiovascular diseases. Desynchronization body metabolism contributes development diseases arteriosclerosis thrombosis. involved controlling inflammatory metabolisms, which can influence pathology genes critical maintaining robust relationship diurnal variation system.
