Inflammatory bowel disease (IBD) is a global health problem in which gut microbiota dysbiosis plays an important pathogenic role.However, the current drugs for IBD treatment are far from optimal.Previous researches indicated that parthenolide (PTL) had not only anti-cancer properties but also strong anti-inflammatory activities.Rationale: To investigate protective effect of PTL on colon inflammation and demonstrate underlying microbiota-dependent mechanism.Methods: Colon severity mouse model was measured by body weight change, mortality, length, activity index (DAI) score, H&E staining colonoscopy evaluation.Gut alteration short-chain fatty acids (SCFAs) production were analyzed through 16S rRNA sequencing targeted metabolomics.Luminex cytokine microarray Enzyme-linked immunosorbent assay (ELISA) conducted to measure cytokines profile.The frequency immune cells lamina propria (LP) spleen phenotyped flow cytometry.Results: The PTL-treated mice showed significantly relieved inflammation, as evidenced reduction loss, survival rate, shortening DAI histology score score.Notably, when depleted using antibiotic cocktails, disappeared.PTL downregulated level proinflammatory cytokines, including IL-1β, TNF-α, IL-6, IL-17A upregulated immunosuppressive IL-10 tissue.16S exhibited much more abundant microbial diversity flora composition.Targeted metabolomics analysis manifested increased SCFAs mice.Additionally, administration selectively colonic regulatory T (Treg) well ratio helper type 17 (Th17) cells, improving Treg/Th17 balance maintain intestinal homeostasis.Gut depletion fecal transplantation (FMT) performed confirm this mechanism.Conclusions: ameliorated manner.The mechanism associated with improved mucosa mediated microbiota-derived production.Collectively, our results demonstrated role potential modulator prevent treat IBD.

Abstract Inflammatory bowel diseases (IBD), composed mainly of Crohn’s disease (CD) and ulcerative colitis (UC), are strongly implicated in the development intestinal inflammation lesions. Its exact etiology pathogenesis still undetermined. Recently accumulating evidence supports that group 3 innate lymphoid cells (ILC3) responsible for gastrointestinal mucosal homeostasis through moderate generation IL-22, IL-17, GM-CSF physiological state. ILC3 contribute to progression aggravation IBD while both IL-22 along with IFN-γ, overexpressed by dysregulation NCR − or + function bias towards ILC1 as well regulatory ILC dysfunction pathological Herein, we feature cells’ development, biological function, maintenance gut homeostasis, mediation occurrence, potential application therapy.
