Radiotherapy (RT) uses ionizing radiation to eradicate localized tumors and, in rare cases, control outside of the irradiated fields via stimulating an antitumor immune response (abscopal effect). However, therapeutic effect RT is often limited by inherent physiological barriers tumor microenvironment (TME), such as hypoxia, abnormal vasculature, dense extracellular matrix (ECM), and immunosuppressive TME. Thus, it critical develop new strategies that can remodel TME overcome radio-resistance suppression. In past decade, high-Z-element nanoparticles have been developed increase radiotherapeutic indices reducing X-ray doses side effects normal tissues enhance abscopal activating elicit systemic immunity. this review, principles radiosensitization, mechanisms suppression, use various sensitize TMEs for enhanced efficacy are discussed. The challenges clinical translation multifunctional TME-remodeling nanoradiosensitizers also highlighted.

Transition-metal-based nanozymes emerge promising therapeutic nanodrugs that provide a reference therapy. In this review, we present recent breakthrough First, comprehensively outline preparation strategies involved in creating transition-metal-based nanozymes, including hydrothermal method, solvothermal chemical reduction biomimetic mineralization and sol–gel method. Though immunogenic cell death (ICD) has garnered significant attention in the realm of anticancer therapies, effectively stimulating strong immune responses with minimal side effects deep-seated tumors remains challenging. Herein, we introduce a novel self-assembled near-infrared-light-activated ruthenium(II) metallacycle, Ru1105 (λem = 1105 nm), as first example Ru(II) supramolecular ICD inducer. synergistically potentiates immunomodulatory and reduces adverse through multiple regulated approaches, including NIR-light excitation, increased reactive oxygen species (ROS) generation, selective targeting tumor cells, precision organelle localization, improved penetration/retention capabilities. Specifically, demonstrates excellent depth-activated ROS production (∼1 cm), resistance to diffusion, anti-ROS quenching. Moreover, exhibits promising results cellular uptake generation cancer cells multicellular spheroids. Importantly, induces more efficient an ultralow dose (10 μM) compared conventional agent, oxaliplatin (300 μM). In vivo experiments further confirm Ru1105's potency inducer, eliciting CD8+ T depleting Foxp3+ effects. Our research lays foundation for design secure exceptionally potent metal-based agents immunotherapy.

Radiotherapy is inevitably accompanied by some degree of radiation resistance, which leads to local recurrence and even therapeutic failure. To overcome this limitation, herein, we report the room-temperature synthesis an iodine- ferrocene-loaded covalent organic framework (COF) nanozyme, termed TADI-COF-Fc, for enhancement radiotherapeutic efficacy in treatment radioresistant esophageal cancer. The iodine atoms on COF not only exerted a direct effect radiotherapy, increasing its X-ray absorption, but also promoted radiolysis water, increased production reactive oxygen species (ROS). In addition, ferrocene surface decoration disrupted redox homeostasis levels hydroxyl lipid peroxide radicals depleting intracellular antioxidants. Both vitro vivo experiments substantiated excellent response TADI-COF-Fc. This study demonstrates potential COF-based multinanozymes as radiosensitizers suggests possible integration strategy combination oncotherapy.

Abstract Nanomedicine‐assisted sonodynamic therapy (SDT) has emerged as one of the most promising cancer therapies due to its unique advantages high penetration, non‐radiation, and excellent oxidative stress effect, but always suffered from self‐protection mechanism apoptosis resistance characteristics evolutionarily mutated cells. Regulated cell death (RCD) received increasing attention in precision treatments because significant role synergistically sensitizing reversing immunosuppressive microenvironment during SDT nanomedicine‐triggered immunogenic death.
