Опубликована: Дек. 14, 2022

Targeting cytokines in inflammatory bowel disease (IBD) is a useful clinical approach. Potential therapies for IBD include regulatory T cell transfer to restore cytokine balance, blocking proinflammatory (e.g., IL-12 and IL-23) or their receptors (sIL-6R IL-36R), inhibiting signaling kinases JAK). An emerging trend therapy combine several anti-cytokine agents simultaneously.

Disruption of the intestinal epithelial barrier is a hallmark mucosal inflammation. It increases exposure immune system to luminal microbes, triggering perpetuating inflammatory response. For several decades, stimuli-induced breakdown human gut was studied in vitro by using colon cancer derived cell lines. While providing wealth important data, these lines do not completely mimic morphology and function normal cells (IEC) due cancer-related chromosomal abnormalities oncogenic mutations. In this review, the role of innate and adaptive immunity in pathogenesis inflammatory bowel diseases (IBD) is reported. IBD, an altered often found, with increased Th17 decreased Treg cells infiltrating intestinal mucosa. An associated increase cytokines, such as IL-1 TNF-α, a decrease anti-inflammatory IL-10, concur favoring persistent inflammation gut Autoinflammation highlighted insights inflammasomes, which activation by exogenous or endogenous triggers might be favored mutations NOD NLRP proteins. Autoimmunity mechanisms also take place IBD context immune stimulation bacterial antigens derived from degradation, response takes results antibodies autoantibodies production, frequent finding these diseases. Inflammation, autoinflammation autoimmunity altering mucus layer enhancing permeability, sustains vicious cycle further mucosal inflammation.

In inflammatory bowel disease microenvironment, transdifferentiation of myeloid-derived suppressor cells (MDSCs) and M2 macrophage accumulation are crucial for the transition colitis-to-cancer. New insights into cross-talk underling mechanism between MDSCs during colitis-to-cancer opening new avenues colitis-associated cancer (CAC) prevention treatment. The role underlying that granulocytic (G-MDSCs) or exosomes (Exo) regulates differentiation monocytic (M-MDSCs) macrophages were investigated using immunofluorescence, FACS, IB analysis, etc, employing siRNA antibodies. vivo efficacy mechanistic studies conducted with dextran sulfate sodium-induced CAC mice, employed IL-6 Abs STAT3 inhibitor.G-MDSCs promote M-MDSC through exosomal miR-93-5 p which downregulating activity in M-MDSC. is responsible enrichment G-MDSC (GM-Exo). Mechanistically, chronic inflammation-driven synthesis via IL-6R/JAK/STAT3 pathway. Early use enhances effect inhibitor against CAC.IL-6-driven secretion promotes involves a signaling transition. Combining inhibitors strategies inhibit IL-6-mediated production beneficial treatment CAC.

Mastitis is an inflammatory response in the mammary gland that results huge economic losses breeding industry. The aetiology of mastitis complex, and pathogenesis has not been fully elucidated. It commonly believed induced by pathogen infection induces a local response. However, clinic, often comorbid or secondary to gastric disease, control effects targeting are limited. In addition, recent studies have found gut/rumen microbiota contributes development proposed gut/rumen–mammary axis. Combined with indicating disturbance can damage gut mucosa barrier, bacteria their metabolites migrate distal extraintestinal organs. occurrence related only external pathogenic microorganisms but also gastroenterogennic pathway.
