Nicotinamide (NAM), the amide form of vitamin B3, is a precursor to essential cofactors nicotinamide adenine dinucleotide (NAD⁺) and NADPH. NAD⁺ integral numerous cellular processes, including metabolism regulation, ATP production, mitochondrial respiration, reactive oxygen species (ROS) management, DNA repair, senescence, aging. NAM supplementation has demonstrated efficacy in restoring energy, repairing damage, inhibiting inflammation by suppressing pro-inflammatory cytokines release. Due its natural presence variety foods excellent safety profile—even at high doses up 3 g/day—NAM extensively used chemoprevention non-melanoma skin cancers treatment dermatological conditions such as blistering diseases, atopic dermatitis, rosacea, acne vulgaris. Recently, anti-aging properties have elevated NAM’s prominence skincare formulations. Beyond repair energy replenishment, significantly impacts oxidative stress reduction, cell cycle apoptosis modulation. Despite these multifaceted benefits, comprehensive molecular mechanisms underlying actions remain not fully elucidated. This review consolidates recent research shed light on mechanisms, emphasizing critical role health therapeutic potential. By enhancing our understanding, this work underscores importance continued exploration into applications, aiming inform future clinical practices innovations.

Skin aging is a complex process involving structural and functional changes characterized by decrease in collagen content, reduced skin thickness, dryness, the formation of wrinkles. This underpinned multiple mechanisms including free radical theory, inflammation photoaging metabolic theory. The immune system, an indispensable part body’s defense mechanism, comprises macrophages, lymphocytes, dendritic cells, mast cells. These cells play pivotal role maintaining homeostasis responding to injury or infection. As age advances, along with various internal external environmental stimuli, may undergo senescence accelerated aging, cell division capability, increased mortality, gene expression patterns signaling pathways, altered functions. collectively impact overall function system. review summarizes relationship between immunity explores characteristics composition effects these on aging. Autoimmune thyroid diseases (AITD) are among the most frequent autoimmune disorders, with a multifactorial etiology in which both genetic and environmental determinants probably involved. Celiac disease (CeD) also represents public concern, given its increasing prevalence due to recent improvement of screening programs, leading detection silent subtypes. The two conditions may be closely associated common risk factors, including setting, changes composition diversity gut microbiota, deficiency nutrients like vitamin D. This comprehensive review discussed current evidence on pivotal role D modulating microbiota dysbiosis immune system dysfunction, shedding light possible relevance an adequate intake this nutrient primary prevention AITD CeD. While future technology-based strategies for proper supplementation could attractive context personalized medicine, several issues remain defined, standardized assays determination, timely recommendations functioning, longitudinal studies randomized controlled trials definitely establish causal relationship between serum levels onset
