In Ethiopia, medicinal herbs have played a prominent role in relieving pain and curing various diseases for humans animals many years. However, consumers don't know what kinds of phytochemicals are the plants or amount metal they contain. Thus, this study, polyphenol content, flavonoid antioxidant capacities, antibacterial activity level metals Asparagus africanus root, Ferula communis, Stephania abyssinia, Brucea antidysenterica Vernonia amygdalina, were analyzed. Identifying phytochemicals, activities, levels ensures their safety efficacy provides deeper understanding healing properties areas where modern medicines not easily accessible. The was determined by 2,2-diphenyl- 1-picrylhydrazyl (DPPH) radical assay, evaluated disc diffusion method. Among studied plant extracts, amygdalina displayed maximum phenolic content (810 mg GAE/100 g), (515.78 QE/100 DPPH scavenging (14.42 AAE/g), whereas least total (160.82 (30.65 (1.14 AAE/g) recorded africanus. highest inhibition zone recorded, with an inhibitory 11 mm S. aurous E. coli. obtained gram-negative bacteria (K. pneumonia) diameter less than 8 mm. contained Mn Fe, while Cu Zn found to be abyssinia. The rising interest in bioactive phyto-compounds, known for their antioxidant, phenolic, and flavonoid properties, has focused attention on Moringa oleifera. With its abundant phyto-molecules, is a promising source biomolecule extraction. As significant vegetable crop, provides ample nutrients that can be efficiently extracted using novel methodologies. This study aims to optimize ultrasound-assisted extraction from leaves, considering diverse parameter combinations improved yield quality. Ultrasound-assisted extraction, notable advancement, considers parameters like amplitude, temperature, time. Employing Response Surface Methodology (RSM) with 20 simulated runs, this investigation assessed yield, Total Phenol Content (TPC), Flavonoid (TFC), Radical Scavenging Activity (RSA). optimized technique yielded an of 12.78-23.28%, TPC 100.12-149.22 mg GAE/g, TFC 14.43-20.19 QE/g, RSA 58.87-85.08%.
