Pioneer transcription factors have the unique and important role of unmasking chromatin domains during development to allow implementation new cellular programs. Compared with those other factors, this activity implies that pioneer can recognize their target DNA sequences in so-called compacted or "closed" heterochromatin trigger remodeling adjoining landscape provide accessibility nonpioneer factors. Recent studies identified several steps action, namely rapid but weak initial binding stabilization followed by opening loss cytosine-phosphate-guanine (CpG) methylation provides epigenetic memory. Whereas CpG demethylation depends on replication, does not. In Minireview, we highlight properties factor class challenges understanding mechanism action.
