Objective To explore the influence of serum metabolites on risk psoriasis. Methods In initial stage, we applied Mendelian randomization to evaluate association between 1,400 and Causal effects were primarily assessed through Inverse-Variance Weighted method Wald Ratio’s odds ratios, 95% confidence intervals. False Discovery Rate was used for multiple comparison corrections. Sensitivity analyses conducted using Cochran’s Q Test, MR-PRESSO. MR-Steiger Test employed check reverse causality. validation sought other sources psoriasis GWAS data verify results meta-analysis combine effect sizes obtain robust causal relationships. addition, also metabolic pathway enrichment analysis known that have a relationship with in both stages. Results identified 112 causally associated psoriasis, including 32 metabolite ratios 80 (69 11 unknown). 24 (16 known, 1 unknown, 7 ratios) confirmed onset. Meta-analysis showed overall combined consistent main direction Of 16 metabolites, most attributed lipid metabolism, 5 as factors 8 protective Peptidic Gamma-glutamylvaline levels had negative while exogenous Catechol sulfate amino acid 3-methylglutaconate positive disease The two stages are mainly enriched following pathways: Glutathione Alpha Linolenic Acid Linoleic Metabolism, Biosynthesis unsaturated fatty acids, Arachidonic Glycerophospholipid metabolism. Conclusion Circulating may potential risk, targeting specific benefit diagnosis, assessment, treatment.

Abstract Background Skin cutaneous melanoma (SKCM) is an aggressive form of malignant with poor prognosis and high mortality rates. Disulfidptosis a newly discovered cell death regulatory mechanism caused by the abnormal accumulation disulfides. This unique pathway guiding significant new research to understand cancer progression for targeted treatment. However, correlation between disulfidptosis long non‐coding RNAs (lncRNAs) in SKCM remains unknown at present. Methods The Cancer Genome Atlas database furnished lncRNA expression data clinical information patients. Pearson Cox regression analyses identified disulfidptosis‐related lncRNAs associated prognosis. ROC curves nomogram validated model. TME, immune infiltration, GSEA analysis, checkpoint gene profiling, drug sensitivity were assessed low‐risk groups. Consistent clustering categorized patients personalized treatment guidance. Results A total twelve development prediction models. area under curve (AUC) values provided reliable discrimination evaluate prognostic potential TME played crucial role tumorigenesis, prognosis, risk scores closely related infiltration. Meanwhile, combination chemotherapy, therapy, immunotherapy was recommended based on efficacy analyses. Conclusion We model that could be used predict help guide chemotherapy plans.

Abstract Background As the most important modifications on RNA level, N6‐methyladenosine (m6A‐) and 5‐methylcytosine (m5C‐) modification could have a direct influence RNAs. Long non‐coding RNAs (lncRNAs) also be modified by methylcytosine modification. Compared with mRNAs, function of lncRNAs more potent to some extent in biological processes like tumorigenesis. Until now, rare reports been done associated cutaneous melanoma. Herein, we wonder if m6A‐ m5C‐ immune landscape prognosis melanoma, want find which directly affect malignant behaviors Methods Systematically, explored expression pattern melanoma from datasets including UCSC Xena NCBI GEO, prognostic were selected. Then, according lncRNAs, samples these divided into several subtypes. Prognostic model, nomogram survival drug sensitivity, GO, KEGG pathway analysis performed.
