Abstract Cellular senescence is a key contributor to aging and aging-related diseases, but its metabolic profiles are not well understood. Here, we performed systematic analysis of the features four types cellular (replication, irradiation, reactive oxygen species [ROS], oncogene) in 12 cell lines using genome-wide modeling meta-analysis. We discovered that replicative ROS-induced share common signature, marked by decreased lipid metabolism downregulated mevalonate pathway, while irradiation oncogene-induced exhibit more heterogeneity divergence. Our knockout simulations showed enhancing administrating for instance, could reverse alterations associated with human tissue aging, suggesting potential anti-aging or lifespan-extending effect. Indeed, experiment Caenorhabditis elegans significantly increased lifespan. study provides new insight into landscape identifies targets interventions.

Cardiac aging constitutes a significant risk factor for cardiovascular diseases prevalent among the elderly population. Urgent attention is required to prioritize preventive and management strategies age-related conditions safeguard well-being of individuals. In response this critical challenge, Aging Biomarker Consortium (ABC) China has formulated an expert consensus on cardiac biomarkers. This draws upon latest scientific literature clinical expertise provide comprehensive assessment biomarkers associated with aging. Furthermore, it presents standardized methodology characterizing across three dimensions: functional, structural, humoral. The functional dimension encompasses broad spectrum markers that reflect diastolic systolic functions, sinus node pacing, neuroendocrine secretion, coronary microcirculation, metabolism. structural domain emphasizes imaging relevant concentric remodeling, artery calcification, epicardial fat deposition. humoral aspect underscores various systemic (N) heart-specific (X) markers, including endocrine hormones, cytokines, other plasma metabolites. ABC's primary objective establish robust foundation assessing aging, thereby furnishing dependable reference applications future research endeavors. aims contribute significantly enhancement health overall

As the global demographic landscape continues to shift towards an aged population, so does medical and socioeconomic burden of cardiovascular diseases. Indeed, ageing is one of, if not the, key risk factor for development However, there are currently no approved therapeutics that primarily target molecular cellular mechanisms underlying process itself. In this review, we present potential emerging anti-ageing strategies, including epigenetic rejuvenation, metabolic reprogramming, autophagy activation, as well senolytic anti-inflammatory therapies, in delaying or reversing age-related disorders, while considering sex differences. doing so, implicate processes pathogenesis several prevalent diseases, such atherosclerosis, hypertension, various types cardiomyopathies (including its hypertrophic, ischemic, dilated, diabetic, arrhythmogenic forms) heart failure, particularly with preserved ejection fraction. Finally, outline future challenges steps needed implementation these novel strategies clinical setting, aim challenging long-held notion a 'nonmodifiable'

Cellular senescence (CS), classically considered a stable cell cycle withdrawal, is hallmarked by progressive decrease in growth, differentiation, and biological activities. Senescent cells (SNCs) display complicated senescence-associated secretory phenotype (SASP), encompassing variety of pro-inflammatory factors that exert influence on the biology both surrounding tissue. Among global mortality causes, cardiovascular diseases (CVDs) stand out, significantly impacting living quality functional abilities patients. Recent data suggest accumulation SNCs aged or diseased systems, suggesting their potential role impairing function. CS operates as double-edged sword: while it can stimulate restoration organs under physiological conditions, also participate organ tissue dysfunction pave way for multiple chronic pathological states. This review explores mechanisms underlie delves into distinctive features characterize SNCs. Furthermore, we describe involvement progression CVDs. Finally, study provides summary emerging interventions either promote suppress discusses therapeutic

Multidrug-resistant sepsis (MDR) is a pressing concern in intensive care unit (ICU) settings, specifically among geriatric patients who experience age-related immune system changes and comorbidities. The aim of this review to explore the clinical impact MDR ICU shed light on healthcare challenges associated with its management. We conducted comprehensive literature search using National Center for Biotechnology Information (NCBI) Google Scholar engines. Our incorporated keywords such as "multidrug-resistant sepsis" OR "MDR sepsis", "geriatric patients" "elderly patients", "complications", "healthcare burdens", "diagnostic challenges", challenges" "ICU "geriatric/elderly patients". This explores specific risk factors contributing sepsis, complexities diagnostic challenges, burden faced by elderly patients. Notably, population bears higher (57.5%), influenced various factors, including comorbidities, immunosuppression, changes, resource-limited settings. Furthermore, imposes significant economic systems, annual costs exceeding $27 billion USA. These findings underscore urgency addressing need tailored interventions improve outcomes reduce costs.
