Neurodegenerative diseases are common, incurable neurological disorders with high prevalence, and lead to memory, movement, language, intelligence impairments, threatening the lives health of patients worldwide. The blood–brain barrier (BBB), a physiological between central nervous system peripheral blood circulation, plays an important role in maintaining homeostasis intracerebral environment by strictly regulating transport substances brain. Therefore, it is difficult for therapeutic drugs penetrate BBB reach brain, this affects their efficacy. Nanoparticles (NPs) can be used as drug carriers also known nanoparticle-based delivery systems (NDDSs). These not only increase stability but facilitate crossing through improve In article, we provided overview types administration routes NPs, highlighted preclinical clinical studies NDDSs neurodegenerative diseases, summarized combined strategies management diseases. Finally, prospects challenges recent basic research were discussed. Above all, provide inspiring strategy treatment

Mitochondria and mitochondria-mediated signalling pathways are known to control synaptic signalling, as well long-lasting changes in neuronal structure function. Mitochondrial impairment is linked dysfunction normal ageing age-associated neurodegenerative ailments, including Parkinson's disease (PD) Alzheimer's (AD). Both proteolysis mitophagy perform a major role neuroprotection, by maintaining healthy mitochondrial population during ageing. Mitophagy, highly evolutionarily conserved cellular process, helps the clearance of damaged mitochondria thereby maintains metabolic balance, energy supply, survival health. Besides maintenance brain homeostasis, hippocampal also synapse formation, axonal development, dopamine release long-term depression. In contrast, defective contributes age-related neurodegeneration promoting accumulation leading dysfunction. Exercise, stress management, dynamics administering natural or synthetic pharmacological compounds some strategies used for neuroprotection neurological diseases. The current review discusses impact conditions, underlying molecular potential therapies based on recently elucidated mitophagy-inducing strategies.

Abstract Quercetin is a flavonoid with notable pharmacological effects and promising therapeutic potential. plays significant role in neuroinflammation, which helps reduce Alzheimer's disease (AD) severity. (Q) quercetin 3-O-glucuronide (Q3OG) are some of the most potent antioxidants available from natural sources. However, form converted into Q3OG when reacted intestinal microbes. The study aims to ensure potential Q Q3OG. In this study, molecular targets were first identified using Swiss Target Prediction platform pathogenic AD DisGeNET database. Followed by compound target overlapping, 77 placed that AKT1, EGFR, MMP9, TNF, PTGS2, MMP2, IGF1R, MCL1, MET PARP1 was top-ranked target, estimated CytoHubba plug-in. Molecular docking performed for towards PDB:1UNQ target. binding score − 6.2 kcal/mol 6.58 respectively. dynamics simulation conducted at 200 ns. This study's results help identify multiple sites bioactive compounds. Thus, synthesizing new chemical entity-based on structural modification may aid eradicating complications.

Metals serve important roles in the human body, including maintenance of cell structure and regulation gene expression, antioxidant response, neurotransmission. High metal uptake nervous system is harmful because it can cause oxidative stress, disrupt mitochondrial function, impair activity various enzymes. Metal accumulation lifelong deterioration, severe neurological problems. There a strong association between accidental exposure neurodegenerative disorders, Alzheimer’s disease (AD), most common form dementia that causes degeneration aged. Chronic to metals well-known environmental risk factor has become more widespread due rapid pace at which activities are releasing large amounts into environment. Consequently, humans exposed both biometals heavy metals, affecting homeostasis molecular biological levels. This review highlights how these affect brain physiology immunity their creating proteins such as β-amyloid tau AD.
