Trimethylamine N-oxide (TMAO), a metabolite, is co-metabolite produced by both gut microbiota and livers, originating from foods rich in choline or carnitine. Abstract Diet has emerged as a key impact factor for gut microbiota function. However, the complexity of dietary components makes it difficult to predict specific outcomes. Here we investigate plant-derived nanoparticles (PNP) on and metabolites in context cancer immunotherapy with humanized gnotobiotic mouse model. Specifically, show that ginger-derived exosome-like nanoparticle (GELN) preferentially taken up by Lachnospiraceae Lactobacillaceae mediated digalactosyldiacylglycerol (DGDG) glycine, respectively. We further demonstrate GELN aly-miR159a-3p enhances anti-PD-L1 therapy melanoma inhibiting expression recipient bacterial phospholipase C (PLC) increases accumulation docosahexaenoic acid (DHA). An increased level circulating DHA inhibits PD-L1 tumor cells binding promoter subsequently prevents c-myc-initiated transcription PD-L1. Colonization germ-free male mice bacteria from non-responding patients supplemented efficacy compared controls. Our findings reveal previously unknown mechanistic PNP human modulating metabolic pathways.

Introduction Vascular smooth muscle cells (VSMCs) are the predominant cell type in medial layer of aorta, which plays a critical role aortic diseases. Innate immunity is main driving force for cardiovascular Methods To determine roles innate VSMC and pathologies, we performed transcriptome analyses on aortas from ApoE –/– angiotensin II (Ang II)-induced aneurysm (AAA) time course, atherosclerosis as well VSMCs stimulated with danger-associated molecular patterns (DAMPs). Background:Sishen Pills (SSPs) are commonly used to treat diarrhea with kidney-yang deficiency syndrome.Trimethylamine-N-oxide (TMAO) is produced through the metabolism of gut microbiota and can participate in syndrome by mediating "gut-kidney axis" transmit inflammatory factors.This study combined network pharmacology animal experiments explore whether SSPs affecting interaction between TMAO microbiota. Material/Methods:A mouse model was constructed using adenine Folium sennae decoction, SSP decoction for treatment.This utilized predict potential mechanisms treating syndrome.16S rRNA high-throughput sequencing analyze mucosal microbial characteristics.ELISA measure TMAO, NOD-like receptor thermal protein domain associated 3 (NLRP3), interleukin-1b (IL-1b), transforming growth factor-b1 (TGF-b1) levels.We performed Masson immunohistochemical (Occludin, ZO-1) staining kidney small intestinal tissues.The fluorescein diacetate (FDA) hydrolysis spectrophotometric method assess activity contents intestine.
