Epigenetic factors are critical regulators of biological and pathological mechanisms they could interact with different molecular pathways. Targeting epigenetic has been an idea approach in disease therapy, especially cancer. Abstract Tumor biomarkers, the substances which are produced by tumors or body’s responses to during tumorigenesis and progression, have been demonstrated possess critical encouraging value in screening early diagnosis, prognosis prediction, recurrence detection, therapeutic efficacy monitoring of cancers. Over past decades, continuous progress has made exploring discovering novel, sensitive, specific, accurate tumor significantly promoted personalized medicine improved outcomes cancer patients, especially advances molecular biology technologies developed for detection biomarkers. Herein, we summarize discovery development including history conventional innovative used biomarker classification biomarkers based on tissue origins, application clinical management. In particular, highlight recent advancements biomarker-based anticancer-targeted therapies emerging as breakthroughs promising strategies. We also discuss limitations challenges that need be addressed provide insights perspectives turn into opportunities this field. Abstract Tumor immunotherapy exerts its anti-tumor effects by stimulating and enhancing immune responses of the body. It has become another important modality therapy with significant clinical efficacy advantages compared to chemotherapy, radiotherapy targeted therapy. Although various kinds tumor immunotherapeutic drugs have emerged, challenges faced in delivery these drugs, such as poor permeability low cell uptake rate, had prevented their widespread application. Recently, nanomaterials emerged a means for treatment different diseases due targeting properties, biocompatibility functionalities. Moreover, possess characteristics that overcome defects traditional immunotherapy, large drug loading capacity, precise easy modification, thus leading wide application immunotherapy. There are two main classes novel nanoparticles mentioned this review: organic (polymeric nanomaterials, liposomes lipid nanoparticles) inorganic (non-metallic metallic nanomaterials). Besides, fabrication method nanoparticles, Nanoemulsions, was also introduced. Abstract Metastasis accounts for 90% of cancer-related deaths among the patients. The transformation epithelial cells into mesenchymal with molecular alterations can occur during epithelial–mesenchymal transition (EMT). EMT mechanism accelerates cancer metastasis and drug resistance ability in human cancers. Among different regulators EMT, Wnt/β-catenin axis has been emerged as a versatile modulator. Wnt is active form physiological condition due to function GSK-3β that destructs β-catenin, while ligand–receptor interaction impairs increase β-catenin stability promote its nuclear transfer. Regarding oncogenic Wnt/β-catenin, upregulation occurs cancers it accelerate EMT-mediated resistance. stimulation by binding ligands Frizzled receptors enhance accumulation cytoplasm stimulates related genes upon translocation. Wnt/β-catenin/EMT implicated augmenting both solid hematological tumors. Wnt/EMT-mediated promotes malignant behavior tumor cells, causing therapy be modulated upstream mediators which non-coding RNAs are main regulators. Abstract Exosomal circRNA serves a novel genetic information molecule, facilitating communication between tumor cells and microenvironmental cells, such as immune fibroblasts, other components, thereby regulating critical aspects of cancer progression including escape, angiogenesis, metabolism, drug resistance, proliferation metastasis. Interestingly, microenvironment have new findings in influencing escape mediated by the release exosomal circRNA. Given intrinsic stability, abundance, broad distribution circRNAs, they represent excellent diagnostic prognostic biomarkers for liquid biopsy. Moreover, artificially synthesized circRNAs may open up possibilities therapy, potentially bolstered nanoparticles or plant exosome delivery strategies. In this review, we summarize functions underlying mechanisms cell non-tumor cell-derived progression, with special focus on their roles immunity metabolism. Finally, examine potential application therapeutic targets, highlighting promise clinical use.
