Curcumin, a polyphenolic compound derived from the turmeric plant (Curcuma longa), has garnered significant attention for its potential anticancer properties due to ability modulate multiple molecular targets involved in cancer development and progression. This chapter provides comprehensive overview of curcumin’s mechanisms action therapy, focusing on effects cell proliferation, apoptosis, metastasis, inflammation. Curcumin exerts through various signaling pathways, including suppression NF-κB, JAK/STAT3, PI3K/Akt, modulation cycle regulators. Despite promising potential, curcumin faces challenges clinical applications poor bioavailability, low solubility, rapid metabolism. To overcome these limitations, innovative drug delivery systems, such as nanoparticles, liposomes, polymer-based formulations, have been developed enhance therapeutic efficacy stability. These systems improve targeted delivery, controlled release curcumin, thereby maximizing while minimizing systemic toxicity. Moreover, preclinical studies assessing combination with other chemotherapeutic agents role an adjuvant treatment shown results. The concludes by discussing future directions curcumin-based therapies, need further trials establish optimal dosage, formulation, regimens be integrated into personalized treatments.

Abstract Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by the accumulation of amyloid‐beta plaques and neurofibrillary tangles, leading to neuronal loss. Curcumin, polyphenolic compound derived from Curcuma longa , has shown potential neuroprotective effects due its anti‐inflammatory antioxidant properties. This review aims synthesize current preclinical data on anti‐neuroinflammatory mechanisms curcumin in context AD, addressing pharmacokinetics, bioavailability, as therapeutic adjunct. An exhaustive literature search was conducted, focusing recent studies within last 10 years related curcumin's impact neuroinflammation role AD. The methodology included sourcing articles specialized databases using specific medical subject headings terms ensure precision relevance. Curcumin demonstrates significant properties modulating neuroinflammatory pathways, scavenging reactive oxygen species, inhibiting production pro‐inflammatory cytokines. Despite potential, challenges remain regarding limited bioavailability scarcity comprehensive human clinical trials. emerges promising adjunct AD multimodal benefits. However, further research required overcome establish effective dosing regimens subjects. Developing novel delivery systems formulations may enhance treatment.

Curcumin is a natural compound found in turmeric that exhibits diverse biological activities. However, its poor bioavailability limits therapeutic application, which has led to the development of various bioavailability-improved formulations. In this methodological study, we analyzed whether systematic reviews on curcumin considered systemic oral formulations when synthesizing evidence from human clinical trials. A total 171 published between 2003 and 2022 were included study. From studies, extracted data study characteristics; type curcumin; methods; reporting regarding bioavailability, funding, conflict interest. Our results show rarely consider variable tested Relevant statistical subgroup and/or sensitivity analyses reported methods only 3.5% 6.4% reviews, respectively. more mentioned their discussion (57%) or conclusion (13%). The detailed analysis suggests there broad recognition product as crucial factor affecting health effects curcumin, not accompanied by adequate synthesis. Therefore, most orally administered should be taken with caution.

Sickle cell disease (SCD) is an inherited hemoglobin disorder marked by red blood sickling, resulting in severe anemia, painful episodes, extensive organ damage, and shortened life expectancy. In SCD, increased iron levels can trigger ferroptosis, a specific type of death characterized reactive oxygen species (ROS) lipid peroxide accumulation, leading to damage impairments. The intricate interplay between iron, inflammation, oxidative stress SCD underscores the necessity thoroughly understanding these processes for development innovative therapeutic strategies. This review highlights importance balancing complex interactions among various factors exploitation knowledge developing novel therapeutics this devastating disease.
