Psoriasis is an immune-related disorder that marked by abnormal thickening of the skin, rapid multiplication keratinocytes, and complex interactions between immune cells affected areas. Although psoriasis cannot currently be cured, drugs can alleviate symptoms regulating homeostasis preventing comorbidities. There are many types to treat psoriasis: small-molecule drugs, including corticosteroids; retinoids; vitamin D analogs; immunosuppressants, such as glucocorticoid ointment, tretinoin cream, methotrexate tablets, etc. Macromolecular biological Certolizumab, Secukinumab, Guselkumab, etc., include monoclonal antibodies target various inflammatory signaling pathways. Compared with traditional therapies offer better targeting lower systemic side effects, but their high costs invasive administration modes constrict widespread use. Spesolimab latest agent used interleukin-36 receptor (IL-36R) approved for market use, which significantly reduces risk general pustular (GPP) flare 84%. Additionally, there several agents interleukin-23/T helper 17 cell pathway have already entered Phase II III clinical trials. At present, first-line therapeutic strategy mild topical administration. Systemic therapy phototherapy preferred treating moderate severe types. However, current completely meet needs. More advanced drug delivery systems optimized effects bioavailability required. Nanocarriers emerging proteins, nucleic acids, cell-based therapies. In this review, we analyze status therapeutics discuss novel diverse well We also summarize effectiveness different strategies.

Psoriasis has been a rising concern for over decade, imposing significant challenges to individuals and society. Traditional topical therapy is non-targeted acts systemically, with associated side effects. This increases the global burden both socially economically. review covers evolution of drug molecules nanotechnology-based approaches treatment psoriasis, chronic inflammatory skin disorder no known etiology. Nanotechnology-based offer promising solutions by reducing effects, providing targeted delivery, protecting from degradation, enhancing retention, controlled release. Researchers have investigated incorporation various conventional non-conventional therapeutic agents into nanocarriers psoriasis treatment. The current understanding disease its using combined novel formulation strategies will reduce duration improve quality life in psoriatic conditions.

Inflammatory skin conditions highly influence the quality of life patients suffering from these disorders. Symptoms include red, itchy and painful lesions, which are visible to rest world, causing stigmatization a significantly lower mental health patients. Treatment options often unsatisfactory, as they suffer either low patient adherence or risk severe side effects. Considering this, there is need for new treatments, notably ways delivering drugs. Stimuli-responsive drug delivery systems able deliver their cargo in response given stimulus are, thus, promising treatment inflammatory conditions. For example, use external stimuli such ultraviolet light, near infrared radiation, alteration magnetic field enables release be precisely controlled space time. On other hand, internal induced by pathological condition, including pH upregulation reactive oxygen species enzymes, can utilized create that specifically target diseased achieve better efficacy safety. In latter context, however, it key importance match trigger mechanism system actual features specific condition. Hence, focus this article placed not only on reviewing stimuli-responsive developed treat conditions, but also critically evaluating context diseases. STATEMENT OF SIGNIFICANCE: Skin diseases affect one-third world's population, lowering patients, who deal with symptoms well due visibility symptoms. Current treatments hampered serious Therefore, more emphasis should developing innovative formulations provide safety hold considerable promise regard, where when needed, reducing adverse effects potentially offering outcomes.

Abstract Microneedles (MNs) have emerged as a highly promising technology for delivering drugs via the skin. They provide several benefits, including high drug bioavailability, non‐invasiveness, painlessness, and safety. Traditional strategies intravenous delivery of anti‐tumor risks systemic toxicity easy development resistance, while MN facilitates precise on‐demand release in local tissues. In addition, by further combining with stimulus‐responsive materials, construction smart MNs can be achieved, which respond to specific physical/chemical stimuli from internal or external environment, thereby improving accuracy tumor treatment reducing surrounding tissues/cells. This review systematically summarizes classification, reaction mechanisms MNs, outlines benefits challenges various types details their application latest progress cancer treatment. Finally, prospects are also discussed, bringing inspiration future precision tumors.
